Ziel ist es, Strategien zur Sicherung der Spermaqualität und Maßnahmen zur Steigerung der Produktionseffizienz in Schweinebesamungsstationen zu entwickeln. In verschiedenen Teilprojekten werden moderne spermatologische Diagnostika etabliert und auf ihr fertilitätsdiagnostisches Potential an Tiermodellen getestet. In vitro-Konservierungseffekte werden bei gelagerten Eberspermien erfasst und Strategien zur Verbesserung der Konservierungsfähigkeit entwickelt. Konzepte zur Qualitätssicherung in der Spermaproduktionskette in Schweinebesamungsstationen werden entwickelt und geprüft.Schwerpunkt des aktuellen Projektjahres ist die Untersuchung des Einfluss' verschiedener Verdünnungsgrade auf die Qualität flüssigkonservierter Eberspermien.

Der Gesundheitsstatus von Tierbeständen, insbesondere das Vorkommen von endemisch verbreiteten Zoonosen und sogenannten Produktionserkrankungen, soll mit Hilfe von Antikörperbestimmungen besser definiert werden als ausschließlich mit klinischen Untersuchungen. Für die Analytik soll eine Miniaturisierung der für die Multiserologie angestrebten Tests und deren Zusammenführung in einem simultanen Testlauf angewandt werden, bei dem nur ein Tropfen Untersuchungsmaterial für die Feststellung der für alle Interessengruppen relevanten Antikörper ausreicht. Damit soll ein Instrument der Prävention und der Früherkennung von Produktionserkrankungen und Zoonosen des Schweines entstehen, das einerseits den heutigen Anforderungen an eine Ressourcen schonende und auf Antibiotika weitgehend verzichtende Tierhaltung und andererseits den Anforderungen an eine verbraucherorientierte Produktion von hochwertigen und gesundheitlich unbedenklichen Lebensmitteln tierischen Ursprungs entspricht.

Project Details:
Research on marine mammals was started in Büsum after the first seal mortality in 1988/89. The aim of the scientists is to study the ecology and physiology of marine mammals and to assess the impact of humans on the animals, their health and their population. The studies focus on the entire German waters of the North and Baltic Seas. The scientific focus is on studies of health (e.g., infectious diseases and zoonoses), behavior, habitat use (e.g., telemetry and acoustic monitoring), population trends (e.g., flight counts), and the effects of anthropogenic interventions on marine mammals. Research projects and monitoring programs include studies of dead and live marine mammals and direct contact with the stranding network and seal hunters. Based on the autopsy of found animals, the quality of the reporting forms submitted by the seal hunters can be verified and further developed.

Within the framework of this project, the previously unrecorded report forms of the seal hunters will be digitized and transferred to a database in order to link them with the data recorded as standard in the dead-find monitoring. This will allow for a more comprehensive and objective assessment of the situation of the native marine mammal population. For this purpose, more complex scientific evaluations can be carried out than before and the resulting results can be used directly by competent authorities for the further development of existing management plans.

Results:

In the annual reports submitted to the client, the data on the number and type of missions are summarized and interpreted. Thus, not only can the effort expended be quantified, but also the extent to which an unusually high number of animals were found can be examined. This allows conclusions to be drawn about the status of the populations of the three native marine mammals, and the addition of additional data allows a more informed interpretation of trends in mortality rates.

Harbor porpoises (Phocoena phocoena), harbor seals (Phoca vitulina) and grey seals (Halichoerus grypus) are native to the North Sea, and in smaller numbers also, in the Baltic Sea. All three species are increasingly at risk. Although they have no natural enemies in their habitat, various factors such as pollution, fishing of the seas, global warming and increasing use of the ecosystem by humans, through shipping or offshore wind turbines, can affect these species. These factors can have serious consequences on the health of these mammals.

In this cooperative project specifically, bone density and bone structure in preparations spanning several decades will be compared, and bones will be examined for trace elements and heavy metals such as mercury, lead and selenium. In addition, changes in the food spectrum will be analyzed and searched for stress markers to see whether environmental conditions have changed over time. Project partners will also check the specimens for pathogens. They will try to detect viruses and categorize the parasites of the three mammalian species. Aim of the project is to work out parameters suitable for assessing the health of our marine mammals over an extended time period.

In addition to TiHo, the Zoological Institute and the Zoological Museum of Hamburg University, the German Marine Museum in Stralsund, the Zoological Institute and Museum at the University of Kiel, Hildesheim University and the National History Museum in Denmark and the Swedish Museum of Natural History are involved in the project.

Infektionserreger mit zoonotischem Potential stellen eine herausragende Bedrohung für die öffentliche Gesundheit dar und erfordern Forschung im Rahmen des so genannten One Health-Konzeptes, in dem neben humanmedizinischen Fragestellungen auch die ursprüngliche Tierspezies als Quelle neuaufkommender Infektionserkrankungen miteinbezogen werden muss. Die schnelle Entwicklung von Vakzinen, insbesondere gegen virale Erkrankungen, erfordert ein eingehendes Verständnis der grundlegenden Pathogenese der jeweiligen Erkrankung, sodass eine Zusammenarbeit von Forschern verschiedenster Disziplinen aus Human- und Veterinärmedizin notwendig ist. Im Rahmen des Projektes Zoonoses Anticipation and Preparedness Initiative (ZAPI) erfolgt eine solche enge Kooperation mit zahlreichen Wissenschaftlern, die das langfristige Ziel verfolgt, eine universelle Plattform für die verzögerungsfreie, antigenetische Charakterisierung von Pathogenen und die Entwicklung von Impfstoffen gegen neuartige Infektionserreger zu etablieren. Drei virale Infektionserreger werden als prototypische Modellpathogene genutzt, um dieses Ziel zu erreichen. Die Forschung konzentriert sich hierbei auf das Rift Valley fever virus (RVFV), das Schmallenberg virus (SBV) sowie das Middle-East Respiratory Syndrome Coronavirus (MERS-CoV). Im Rahmen des Projektes konnten im Institut für Pathologie zunächst erfolgreich immunhistologische Methoden zur Charakterisierung von Immunzellen beim Dromedar, einer wichtigen Spezies in der Epidemiologie von MERS-CoV, etabliert werden. Darüber hinaus werden derzeit Studien zur Virusantigenverteilung, Virusrezeptorverteilung sowie zum Zelltropismus des MERS-CoV im oberen Respirationstrakt von vakzinierten und nicht-vakzinierten Dromedaren durchgeführt. Morphologische, immunhistologische und ultrastrukturelle Untersuchungen an Dromedargewebe sowie mit MERS-CoV infizierten Kleintiermodellen ergänzen die gegenwärtigen Untersuchungen. Untersuchungen zur Immunpathogenese von RVFV in infizierten gentechnisch veränderten Mäusen werden gegenwärtig initiiert.

Neurons in the ventral part of the ventral nucleus of the lateral lemniscus (VNLL) show highly precise temporal discharge patterns with high firing rates and low latencies upon monaural (contralateral) stimulation with amplitude-modulated sounds. Therefore, they are believed to play an important role in the processing of conspecific vocalization such as human speech. In this project we evaluate the hypothesis that the markedly biphasic shape of these neurons' excitatory synaptic currents is a key feature in understanding their response properties. To this end, we will quantitatively describe the synaptic dynamics of the afferent synapses and study the supra-threshold integration of these currents using a combination of electrophysiological recordings and a computational model of these neurons. Finally, we will quantify the information transfer and stimulus discriminability of the response patterns using conductance trains derived from human speech signals in experiments and model.

Das Ziel des Projekts ist, grundlegende Erkenntnisse über die Morphologie und Histologie der männlichen und weiblichen Gonaden des Schweinswals der Nord- und Ostsee zu erlangen, um damit Aussagen über die Spermatogenese und Ovogenese, den Zeitpunkt der Geschlechtsreife, die reproduktionsbiologische Saisonalität und eventuelle Veränderungen der Populationsdynamik über zwei Dekaden (1994-2014) treffen zu können.

Project Details:
Neuronal transplantation of appropriate cells into brain regions involved in the modulation of epileptic seizure activity is an experimental therapeutic approach for difficult-to-treat types of epilepsies. We investigate if biodegradable spider silk is able to anchor grafted cells, thereby avoided unwanted migration to areas outside the target regions within the brain. Goal of this experimental epilepsy research project is the longterm fixation and subsequent integration of grafted cells into the neuronal network of the host brain.

Results:

We successfully established a method for fragmentation of spider silk. First in vivo experiments indicate anchoring of grafted cells within the rat brain. Publication is in preparation.
