Tick-borne encephalitis (TBE) is considered as one of the most important viral tick-borne diseases affecting humans. The disease may cause death or permanent disability due to severe neurological damage. TBE thus represents a considerable public health burden. In recent years, a considerable geographic expansion of TBE has been noted in Germany with the consequence the National Institute of Public Health (Robert-Koch-Institut) declared district in the Northern Germ federal state Lower Saxony as official risk area in the beginning of 2019. However, the usually very small areas of TBE virus circulation (so called natural foci) are still mostly unknown in Northern Germany. This study aims to identify the locations of TBE transmission foci (i.e. the presence of TBE-positive ticks) in Lower Saxony, Northern Germany, by following up on the movement patterns and possible sources of tick exposure of TBE patients. Detection of TBE virus occurrence in Northern Germany will help to prevent clinical cases of disease as it can be expected that public awareness about TBE foci will lead to avoidance of known hot spot areas and increase the people´s willingness to vaccinate.

The area of responsibility of advisors is determined and recommendations are given on how responsible advice can be implemented in practice. A concept and material for further training courses will be developed which, on the one hand, clarify the responsibility of advisors and, on the other hand, also convey the animal ethical aspects of advice.

Results:

The provision of advice to farms plays an important role in establishing animal welfare measures in livestock farming. One of the aims of the project was to identify the interrelationships of responsibility in advisory services. To this end, the areas of responsibility were first determined, e.g. in the design of the advisory process or in the decision and implementation of the advisory content.

As values can influence the guidance process, it is always important for guidance practitioners to be clear about their values, whereby it is also important to differentiate between their own values and those of the employer. Critical reflection is an important tool for this. Significant values were identified during the project.

A framework was also created with regard to animal welfare/animal protection, which shows the responsibility of advisors. In the course of this, it was possible to outline the approaches that animal welfare advice must pursue in order to meet the requirements for good advice and to be able to provide sustainable approaches for greater animal welfare.

When discussing animal welfare/animal protection, there are always conflicts of objectives.

It is very important for the success of an advisory service that these conflicting goals are identified.A decision-making cascade developed in the project is intended to provide a remedy here, which is intended to encourage reflection throughout the advisory activity.

The project developed a training program that has already been successfully tested in practice. It dealt with the thematic blocks entitled "Consultancy in the knowledge system of agriculture", "Fundamentals of animal ethics", "Responsibility and scope for responsibility in consultancy", "Values and requirements for ethically "good" consultancy", "Animal welfare and animal protection in consultancy" and "Conflicting objectives and the decision-making cascade".

The extremely large number of advisors from both official and private advisory services who attended the event showed the great interest in this topic in the run-up to the event and thus also the need to address it among advisors.

The project has thus succeeded in defining the area of responsibility of counselors and providing them with recommendations on how responsible counseling can be implemented. The training program clarifies the responsibility of advisors and highlights the importance of advice for animal welfare and animal protection.

Die Tatsache, einen erfolgreichen Zuchtrüden plötzlich und meist unbemerkt für die Zucht zu verlieren, ist beim Besitzer oftmals mit einer starken emotionalen Belastung, aber auch mit wirtschaftlichen Verlusten verbunden. Zusätzlich dazu stellt es den Tierarzt vor große Probleme: Aufgrund der meist späten Vorstellung des Rüdens ist es äußerst schwierig die auslösende Noxe rückblickend zu identifizieren und eine dementsprechende Behandlung einzuleiten. Azoospermie, das Fehlen von Spermien im Ejakulat, ist mit 34,8% der häufigste Befund bei infertilen, andrologisch geschlechtsgesunden Rüden. Sowohl beim Hund als auch bei anderen Spezies, kommt die testikuläre nicht-obstruktive Form der Azoospermie (NOA) am häufigsten vor. Die Diagnose ist mit hochgradigen histologischen Veränderungen, Infiltration von Immunzellen und dem Arrest der Spermatogenese assoziiert, was auf eine spontane autoimmun-vermittelte Orchitis (AIO) hindeutet. Die Art der beteiligten Immunzellen wurde von der AG identifiziert und die Expression von Entzündungsmarkern näher charakterisiert. Bei bereits eingetretener Infertilität ist von einer schlechten Prognose und irreversiblen Schädigung des Gewebes auszugehen. Eine Möglichkeit der Therapie oder Heilung der Erkrankung besteht bisher nicht. Grundvoraussetzung für die Bildung von gesunden Spermien im Ejakulat ist das Vorhandensein der sogenannten spermatogonial stem cells (SCCs) im Hoden. Diese Stammzellpopulation besitzt einerseits die Fähigkeit zur Selbsterneuerung und produziert andererseits sich differenzierende Tochterzellen. Aktuelle Studien beschäftigen sich mit der Verwendung von SSCs zur Behandlung von Infertilität und Fruchtbarkeitsstörungen in der Humanmedizin. Um diese Erkenntnisse auf der Rüden übertragen zu können, ist ein erster essenzieller Schritt zur Behandlung der AIO die Suche nach SSCs im entzündlich veränderten Hodengewebe.

Humane Q-Fieber Erkrankungen werden durch das Bakterium Coxiella burnetii hervorgerufen. In Deutschland gehen Infektionen beim Menschen meist von infizierten Schafen während der Lammzeit aus. Betroffene Tiere scheiden bei der Geburt große Mengen an Coxiellen mit dem Fruchtwasser und der Nachgeburt aus. Durch das Einatmen erregerhaltiger Tröpfchen oder Stäube können sich auch Menschen infizieren, selbst wenn sie keinen direkten Kontakt mit den Tieren haben. Infizierte Schafe sind häufig symptomlos, bei Rind und Ziege kann die Infektion zu Aborten und Reproduktionsstörungen führen. An Q-Fieber erkrankte Personen leiden unter Fieber, Kopfschmerzen und anderen unspezifischen Symptomen. Im weiteren Verlauf kann es zu einer Lungenentzündung und in seltenen Fällen sogar zu einer Herzmuskelentzündung kommen. Bundesweit treten immer wieder örtlich begrenzte Epidemien auf.
Im Rahmen des Q-GAPS-Zoonoseverbundes werden im Teilprojekt (TP) 1 epidemiologische Studien zur Verbreitung von Coxiella burnetii in Herden kleiner Wiederkäuer durchgeführt. Scheiden- und Vorhauttupfer von Schafen und Ziegen werden mittels PCR auf erregerspezifische Gensequenzen untersucht. Diese molekularbiologische Methode identifiziert das Bakterium, bzw. Teile des Bakteriums und gibt auch Hinweise auf die Intensität der Erregerausscheidung. Parallel dazu werden auch Blutproben dieser Tiere auf Antikörper gegen Coxiella burnetii getestet. Weitere Studien beschäftigen sich mit der Ausscheidung des Erregers in milchproduzierenden Schaf- und Ziegenherden (Longitudinalstudie), den wirtschaftlichen Auswirkungen einer Coxiellen-Infektion speziell in Schafherden (Kohortenstudie) und den Effekten einer Impfung auf die Reproduktionsleistung sowie die Erregerausscheidung (Interventionsstudie) in betroffenen Herden.
Im TP 2 wird ein konzeptionelles Design für ein aktives Monitoring- und Surveillance-System (MOSS) zur Detektion und Charakterisierung von C. burnetii in Herden kleiner Wiederkäuer in Deutschland erstellt und die beste Kombination von Detektionsmethoden und Probenmatrizes für C. burnetii in Herden kleiner Wiederkäuer in Deutschland identifiziert. Außerdem werden die Anwendbarkeit eines angepassten latenten Klasse-Regressionsmodells auf diagnostische Testergebnisse von multiplen Matrizes sowie die Anwendbarkeit eines angepassten latenten Transitionsmodells auf die perpetuierte Ausscheidung von C. burnetii in Herden kleiner Wiederkäuer evaluiert. Des Weiteren wird eine Stakeholderanalyse zu den Bedürfnissen und Erwartungen von Human- und Veterinärmedizinern zu Informationen über C. burnetii durchgeführt. Gemeinsam mit Projektpartnern aus Q-GAPS werden das Risiko für das chronische Müdigkeitssyndrom nach einem Q-Fieberausbruch in der menschlichen Bevölkerung analysiert und eine Blaupause zur übergreifenden sekundären Analyse von Meldedaten von Ausbrüchen in Tierpopulationen und der menschlichen Bevölkerung erstellt.

Raw cured products are popular meat products that often get their color from the addition of nitrite during aging. Here, the nitrite reacts with the myoglobin within the muscles to form red nitrosomyoglobin. In addition, the nitrite is also used to suppress the growth of undesirable microorganisms, particularly Clostridium botulinum and Listeria monocytogenes. However, free nitrite may still be present in the meat product, which can react with amines to form nitroso compounds that are harmful to health.
However, Parma ham has been produced for a long time without the addition of nitrite or nitrate, using only salt. The red color of the ham is due to the formation of zinc protoporphyrin (ZnP), which is increasingly formed during the storage of the ham for up to 15 months. In this process, iron is replaced by zinc in myoglobin, regardless of the microbiological contamination of the meat. In the study, raw hams with and without the addition of nitrite are to be produced from turkey breast muscles and pork hams. At various times over a 15-month period, starting from the raw material, meat texture parameters will be examined and microbial status determined at regular intervals throughout the aging period. After maturing, the hams are sliced, subjected to a two-week storage test, analyzed microbiologically and physicochemically and evaluated with regard to its sensory characteristics. The aim of this research project is to verify the feasibility of the production and to determine the quality of an air-dried raw ham made from turkey or pork without nitrite curing salt in comparison to raw hams containing nitrite.

Vom zentralen Nervensystem absteigende inhibitorische Schmerzbahnen sind ein Teil des körpereigenen Systems zur Schmerzmodulation. Wird dieses System stimuliert, werden nachfolgende, schmerzhafte Reize als weniger schmerzhaft wahrgenommen, im englischen Sprachgebrauch wird das als "Diffuse noxious inhibitory control (DNIC)" bezeichnet. In der humanmedizinischen Forschung wird zur Evaluation der DNIC ein standardisierter, konditionierender Stimulus (CPM= conditioned pain modulation) eingesetzt, bevor der eigentliche Teststimulus erfolgt. Ist die DNIC intakt, wird der Teststimulus nach CPM als weniger schmerzhaft als vor/ ohne CPM wahrgenommen.
Für Labornager und in der Humanmedizin konnte gezeigt werden, dass die DNIC bei Individuen mit chronischen Schmerzen weniger stark ausgeprägt eiterhin ist für die Humanmedizin beschrieben, dass Patienten, die vor einer Operation niedrigere Werte im DNIC-Test aufweisen ein erhöhtes Risiko besitzen postoperativ an chronischen Schmerzen zu leiden
Für den Hund ist eine Technik zur Evaluierung der DNIC beschrieben. Eine systematische Evaluierung dieser Testmethodik, bezüglich Zuverlässigkeit und Reproduzierbarkeit beim Hund wurde bisher allerdings nicht durchgeführt.
Die Überprüfung der DNIC kann zu einer besseren Typisierung des vorliegenden Schmerzphenotypes und damit zu einer besseren Anpassung einer effektiven Schmerztherapie beitragen.

Im Rahmen dieses Forschungsprojektes soll eine standardisierte Methode zur Untersuchung der absteigenden inhibitorischen Schmerzbahnen beim Hund erarbeitet und kritisch evaluiert werden.

The preservation of food with chemical or physical methods should reduce the microbial contamination of food and should extend the shelf life of the products in order to minimise the health risk for the consumer. In a previous project at the Institute of Food Quality and Food Safety, the bacteria-reducing effects of Ethyl-Nα-lauryl-L-arginate hydrochloride (LAE) have already been successfully investigated. However, few studies are available that have examined the influence of LAE in combination with a biological preservation method (starter bacteria cultures) on the microbiological contamination with pathogenic and spoilage bacteria as well as on the chemical and sensory quality characteristics of the treated products. These investigations are useful to improve an individual effect by a combined treatment and to check whether these processes change qualitative properties of the food. For this reason, in the present study fresh meat from pigs, cattle and chickens, after inoculation with defined microorganisms, will be individually treated with LAE and starter culture mixtures either alone, or in combination, in order to show the effects of a combined treatment in comparison to the untreated control sample and to the individual treatment. Not only the short-term effects directly after treatment, but also the long-term effects during storage of the products until the usual best-before or use-by date will be investigated.
