Mosquito-borne arboviruses are an increasing (re-)emerging threat for more than 781 million people in Africa (70% of the continental population) (Weetman et al., 2018). Aedes-borne yellow fever (YFV), dengue (DENV), chikungunya (CHIKV) and Zika virus (ZIKV) as well as Aedes- and Culex-borne Rift Valley fever (RVFV) virus infections have been documented from the African continent, but reports are sketchy. The absence of evidence-based risk maps for arbovirus infections prevents adequate planning of both public health and research resources. Although the main vectors Aedes aegypti, Aedes albopitus and Culex quinquefasciatus are prevalent in Central and West African countries (Kreamer et al., 2015); the distribution and the frequency of arboviral infections and outbreaks display a surprising heterogeneity between and within certain countries. Thus, we ask the key question: Do genetic variability and bionomic differences in local mosquito vector populations influence arbovirus occurrence across countries in West Africa?
Here we propose to assess the difference in human exposure to arboviruses (through an epidemiologic and serologic study) based on in-depth multi-national investigation on the vector?s biting behavior, virus surveillance, and the genetic structure of vector populations. We will conduct our study in three West and Central African countries (Benin, Gabon and Côte d?Ivoire) with different and specific epidemiological profiles. Benin (West Africa) will present a setting with low arboviral occurrence, Gabon (Central Africa) will represent a latent focus with high arboviral occurrence where no outbreaks have been reported since 2010, and Côte d?Ivoire (West Africa) will represent an active focus with high arboviral occurrence where ongoing outbreaks have been reported recently. In each country, the study will be conducted in already known arbovirus foci or potential arbovirus foci with arbovirus detection from either human, animal or mosquitoes. As arboviruses have been reported mainly in large African cities, the study will be conducted in the capital cities (and their surrounding towns) of every country to maximize the probability to observe an arboviral infection We will select three sites representing each rural, suburban or urban area to capture potential variability in arbovirus prevalence in connection with mosquito species diversity, and vector and human populations.
The outcomes will help directly to set up a sustainable network on arboviruses (termed EcoVir-network) which will work to contribute to improving the surveillance, prevention and control of arboviruses in Africa. Furthermore, we will build a well-trained new generation of young African scientists through our training program, who will in future contribute to arbovirus control in Africa.

Im Rahmen dieses Projektes wird die Rolle des Makrophagen-induzierbaren C-Typ Lektinrezeptors (MINCLE) im Verlauf der Strongyloides ratti-Infektion untersucht, mit Fokus auf wurmvermittelte Immunmodulation. Ziel des Projektes ist es, die Rolle von MINCLE während der Infektion mit S. ratti im Mausmodell genauer zu untersuchen und Liganden für MINCLE in den Lysaten von S. ratti zu identifizieren und molekular zu charakterisieren. Zusammenfassend erwarten wir von dem Projekt ein genaueres Verständnis der Immunevasion im Verlauf von Wurminfektionen und des Einflusses des C-Typ Lektinrezeptors MINCLE auf diesen Prozess.

Project Details:
Etwa 3 % der Weltbevölkerung ist chronisch mit dem Hepatitis-C-Virus (HCV) infiziert, welches akute und chronische Lebererkrankungen verursacht. Da ein robustes immunkompetentes Tiermodell zur Entwicklung prophylaktischer oder therapeutischer Impfstoffe noch nicht gefunden werden konnte und der Ursprung des HCV schwer zu fassen ist, rückt das dem HCV am nächsten verwandte Virus, das equine Hepacivirus (EqHV) bei Pferden, mehr in den Fokus. Ein verbessertes Verständnis dieses Viruses ist außerdem für die Pferdemedizin von großer Bedeutung, da gezeigt werden konnte, dass auch EqHV akute und chronische Infektionen verursachen kann.
Grund für diese Ausprägung kann eine fehlende hepatische Toleranz sein, die verhindert, dass dauerhaft infizierte Pferde eine robuste zelluläre Immunität aufbauen können. Daher würde ein erweitertes Wissen über die Mechanismen der hepatischen Toleranz helfen, chronischen Infektionen entgegenzuwirken. Hierauf soll die therapeutische Impfung eine Antwort geben, bei der die MVA basierte Vektor-Impfstoff-Technologie verwendet wird. Dabei exprimiert MVA (modified vaccinia virus Ankara) ausgewählte EqHV-Antigene, welche dann in Impfungen vergleichend analysiert werden, um diejenigen Antigene zu finden, die eine ausgewogene zelluläre und humorale antivirale Immunantwort induzieren.
Ziel ist es hierbei, die Immunantwort gegen EqHV bei chronisch infizierten Pferden und die immunologische Wirkung einer therapeutischen Impfung gegen EqHV bei gesunden Tieren zu untersuchen. Darüber hinaus stellt dieses Projekt auch einen potenziellen Nutzen für die Humanmedizin dar und liefert weitere Hinweise auf den Ursprung von HCV.

The aim of this project is to ensure animal welfare when slaughtering large fish. To this end, the effectiveness of stunning these fish with the method of percussion
(knocking off) or non-penetrating and penetrating captive bolt evaluated under aspects of animal welfare. Procedure descriptions for the stunning of sturgeons and catfish are to be drawn up, which will make it possible to include these stunning procedures as permissible procedures for the stunning of fish in the Animal Welfare Slaughter Ordinance. Furthermore, training material is to be made available for plant managers and authorities.

The overall objective of KontRed is to reduce the load of zoonotic pathogens (main-ly Salmonella and Campylobacter) in carcasses at the end of the production chain under hygienic aspects through optimization and control of existing processes as well as through implementing new technical procedures. Concrete Goals of the network are to optimize existing processes during slaughter and processing to reduce occurrence and transfer of zoonotic pathogens. For this purpose, the de-velopment, implementation and validation of biological, chemical, physical and technical control and intervention measures for the reduction of zoonotic patho-gens and the development of an evaluation model to test the efficiency of proce-dures and measures for the reduction of zoonotic pathogens in the slaughter and processing process for poultry and pork will be carried out.

FU Berlin coordinates the network, which involves 17 partners from science and industry. From the University of Veterinary Medicine Hanover two institutes are involved: the Institute for Biometry, Epidemiology and Information Processing (IBEI) and the Institute for Food Quality and Safety (LMQS).

LMQS is working on the development of new biological decontamination strategies. As part of the project, bacteriocins for the reduction of Salmonella and Campylo-bacter in poultry slaughter will be isolated, purified, characterized and tested for their efficacy and stability. Bacteriocins are peptides produced by many bacterial species that have an antibacterial effect on specific other bacterial species. Since the effect of most bacteriocins covers only a narrow host spectrum, they can be used to combat specific bacteria without affecting the entire microbiota.

Core task of the IBEI is to standardize the data collected in the individual subpro-jects and to manage them in a harmonized, specially created database as well as to analyse them descriptively. Furthermore, the institute is involved in the sample planning and data management of primary and secondary data from the individual work packages in consultation with the respective responsible persons. In the fur-ther course of the project, the statistical modelling for the individual sub-aspects will be planned and an overall risk assessment will be carried out in close collabora-tion with the Federal Institute for Risk Assessment (BfR).

Ziel des Projektes ist die weitergehende Entwicklung der Komponenten und der Nachweis der Funktionsweise einer Implantat-dirigierten Anlieferung von Wirkstoffen auf der Basis magnetischer Prinzipien. Auf diesem Wege soll eine neuartige Möglichkeit etabliert werden, spezifische Wirkstoffe mittels magnetischer Nanopartikel bedarfsabhängig zu einem frei wählbaren Zeitpunkt an magnetisierbaren Implantaten zu akkumulieren und somit die Wirksamkeit bei reduzierter Patientenbelastung zu erhöhen. Die Vision des Projektes ist der Einsatz dieser Technik für verschiedene Implantattypen, die gezielt magnetisiert werden, um temporär Wirkstoffe an den Zielort zu verbringen. Damit sollen die Wirksamkeit der Substanzen erhöht, systemische Nebenwirkungen verhindert und die Anzahl von Implantatrevisionen minimiert werden.

Es wird allgemein angenommen, dass virale und bakterielle Co-Infektionen im Respirationstrakt prädisponierend für Infektionen durch Streptococcus suis sind. Wahrscheinlich beruhen die prädisponierenden Effekte auf einer Beeinträchtigung mukoziliärer Clearance- und Barrierefunktionen sowie auf Modulationen der Immunantwort, die Adhärenz und Invasion von S. suis fördern. Allerdings sind die Prozesse und Mechanismen der Interaktionen von S. suis mit co-infizierten respiratorischen Zellen bisher nur wenig bekannt. Unsere bisherigen Ergebnisse ergaben, dass porzine Influenzaviren (SIV) die Interaktionen von S. suis mit respiratorischen Zellen in einem zweistufigen Prozess beeinflussen. Initial wird die Adhärenz und Kolonisation der Streptokokken durch Bindung von Sialinsäureresten der Kapselpolysaccharide an Hämagglutinin an der Oberfläche SIV-infizierter Zellen vermittelt. In einem zweiten Schritt wird durch SIV-induzierte Ziliostase und Reduzierung der mukosalen Barriere die Invasion von S. suis in tiefere Zellschichten ermöglicht. Dieser Prozess ist sehr wichtig für die Pathogenität von S. suis, da er Voraussetzung für die weitere Invasion in den Blutkreislauf und die Ausbreitung des Erregers ist.
Im Folgeprojekt werden wir uns auf die Effekte mukosaler Schäden durch co-infizierende Pathogene auf Interaktionen von S. suis mit der respiratorischen Barriere konzentrieren. Wir gehen davon aus, dass das Ausmaß der mukosalen Schädigung einen entscheidenden Einfluss auf den Verlauf einer S. suis Infektion hat. Wir werden die gleichen primären Zellmodelle nutzen wie im vorherigen Projekt. Allerdings werden wir als co-infizierendes Pathogen statt SIV nun Bordetella bronchiseptica einsetzen. Von B. bronchiseptica ist bekannt, dass in Schweinen Co-Infektionen mit S. suis auftreten (beide sind am Porcine Respiratory Disease Complex beteiligt). Zudem haben experimentelle Infektionen gezeigt, dass B. bronchiseptica Schweine für intranasale Infektionen mit S. suis prädisponiert. Unsere bisherigen Untersuchungen haben außerdem ergeben, dass B. bronchiseptica mukosale Schäden verursacht, die von Ziliostase bis zur Ablösung Zilien-tragender Epithelzellen reichen, was zur Förderung der Adhärenz, Invasion und Zytotoxizität von S. suis führte. Daher sind unsere Hauptziele (i) die Charakterisierung von Effekten (milder und schwerer) mukosaler Schäden durch B. bronchiseptica auf die Adhärenz und Invasion von S. suis, (ii) die nähere Klärung der Rolle des S. suis Toxins Suilysin in diesen Interaktionen sowie (iii) die Analyse der Zellantwort von B. bronchiseptica-infizierten respiratorischen Zellen auf S. suis. Als Erkenntnisgewinn erwarten wir nähere Einblicke in die komplexen Pathogen-Wirt Interaktionen während der initialen Phase von S. suis Infektionen und, allgemein, während bakteriell-bakterieller Co-Infektionen im porzinen Respirationstrakt. Darüber hinaus können unsere Analysen der Wirtszellantworten dazu beitragen, Modulationen mukosaler Immunantworten durch Co-Infektionen zu identifizieren.

The aim of the present study is to evaluate the habitat-improving measures for the target species pheasant and brown hare in the model areas from an ecological perspective, in order to implement them later in a large number of areas if they have positive effects. In this way, the populations of the target species could be stabilised in the long term and, if necessary, increased. Furthermore, the experiences and results of the requested study could contribute to an improvement of the existing agricultural support systems.
It can be assumed that the habitat-improving measures will not only show positive effects on the target species, but will also contribute to an overall increase in biodiversity. Therefore, the research approach outlined here is consistent with previous studies and should result in practical solutions and recommendations for agriculture and hunting that contribute to an improvement in habitat quality for the target species. Furthermore, recommendations for improved predator hunting can also be made from the results.
