Inflammation and associated lung injury, such as the acute respiratory distress syndrome (ARDS) are major causes of morbidity and and mortality in COVID-19. The Nrf2 transcription factor activates a comprehensive cell-protective and anti-inflammatory program. It is known from mouse models that pharmacological activation of Nrf2 and induction of this program protects animals from LPS-induced ARDS. In a cell line-based screen for agents against COVID-19, we found 2 Nrf2 activators that prevent the cytopathic effect of SARS-CoV-2 at nontoxic doses. Therefore, the aims of the present project are,
- the establishment of infection models based on
human respiratory epithelial cells, a model for the alveolar
epithelial cell barrier, and ex vivo culture of lung explants,
- Evaluation of the importance of the endogenous Nrf2 program for
the cellular response against SARS-CoV-2, and
- Verification of the 2 identified Nrf2 activators and
identification of additional protective agents via a
comprehensive screening of known Nrf2 activators.

Humane Q-Fieber Erkrankungen werden durch das Bakterium Coxiella burnetii hervorgerufen. In Deutschland gehen Infektionen beim Menschen meist von infizierten Schafen während der Lammzeit aus. Betroffene Tiere scheiden bei der Geburt große Mengen an Coxiellen mit dem Fruchtwasser und der Nachgeburt aus. Durch das Einatmen erregerhaltiger Tröpfchen oder Stäube können sich auch Menschen infizieren, selbst wenn sie keinen direkten Kontakt mit den Tieren haben. Infizierte Schafe sind häufig symptomlos, bei Rind und Ziege kann die Infektion zu Aborten und Reproduktionsstörungen führen. An Q-Fieber erkrankte Personen leiden unter Fieber, Kopfschmerzen und anderen unspezifischen Symptomen. Im weiteren Verlauf kann es zu einer Lungenentzündung und in seltenen Fällen sogar zu einer Herzmuskelentzündung kommen. Bundesweit treten immer wieder örtlich begrenzte Epidemien auf.
Im Rahmen des Q-GAPS-Zoonoseverbundes werden im Teilprojekt (TP) 1 epidemiologische Studien zur Verbreitung von Coxiella burnetii in Herden kleiner Wiederkäuer durchgeführt. Scheiden- und Vorhauttupfer von Schafen und Ziegen werden mittels PCR auf erregerspezifische Gensequenzen untersucht. Diese molekularbiologische Methode identifiziert das Bakterium, bzw. Teile des Bakteriums und gibt auch Hinweise auf die Intensität der Erregerausscheidung. Parallel dazu werden auch Blutproben dieser Tiere auf Antikörper gegen Coxiella burnetii getestet. Weitere Studien beschäftigen sich mit der Ausscheidung des Erregers in milchproduzierenden Schaf- und Ziegenherden (Longitudinalstudie), den wirtschaftlichen Auswirkungen einer Coxiellen-Infektion speziell in Schafherden (Kohortenstudie) und den Effekten einer Impfung auf die Reproduktionsleistung sowie die Erregerausscheidung (Interventionsstudie) in betroffenen Herden.
Im TP 2 wird ein konzeptionelles Design für ein aktives Monitoring- und Surveillance-System (MOSS) zur Detektion und Charakterisierung von C. burnetii in Herden kleiner Wiederkäuer in Deutschland erstellt und die beste Kombination von Detektionsmethoden und Probenmatrizes für C. burnetii in Herden kleiner Wiederkäuer in Deutschland identifiziert. Außerdem werden die Anwendbarkeit eines angepassten latenten Klasse-Regressionsmodells auf diagnostische Testergebnisse von multiplen Matrizes sowie die Anwendbarkeit eines angepassten latenten Transitionsmodells auf die perpetuierte Ausscheidung von C. burnetii in Herden kleiner Wiederkäuer evaluiert. Des Weiteren wird eine Stakeholderanalyse zu den Bedürfnissen und Erwartungen von Human- und Veterinärmedizinern zu Informationen über C. burnetii durchgeführt. Gemeinsam mit Projektpartnern aus Q-GAPS werden das Risiko für das chronische Müdigkeitssyndrom nach einem Q-Fieberausbruch in der menschlichen Bevölkerung analysiert und eine Blaupause zur übergreifenden sekundären Analyse von Meldedaten von Ausbrüchen in Tierpopulationen und der menschlichen Bevölkerung erstellt.

Holding and managing pigs currently faces diverse adjustments regarding animal welfare, castration, tail docking and housing in crate stands. Farmers discuss this in different blogs and expert forms in the internet. In this study expert opinion in the internet are assessed to evaluate the experts? knowledge regarding the themes named aboved. Citations shall be compared with current evidence base scientific knowledge. Differences shall be described and discussed in order to developed tight-fit training and education.

The persistence of Actinobacillus pleuropneumoniae (App) on the tonsils facilitates the spread of the pathogen in pig populations. Microbiological-cultural detection of the pathogen from tonsils has proven to be difficult because the diverse commensal colonization by other bacteria (e.g., Streptococcus suis) often overgrows App during culturing. The use of selective media leads to higher sensitivity of bacteriological diagnostics, but nevertheless, the success rate compared to molecular biological tests (PCR) is not satisfactory.
In most cases, acute outbreaks of porcine pleuropneumonia occur in persistently infected pigs under the influence of trigger factors. In vitro, the influence of stress hormones, such as cortisol and catecholamines on the growth behavior of the pathogen will be investigated. Possibly, catecholamines and cortisol can be used under in vitro conditions to improve the growth behavior of A. pleuropneumoniae.

Project Details:
The preventive effect of a prebiotic feed on the occurrence of tail biting in pigs is investigated. Situations in which tail biting occurs are characterized in terms of clinical signs, colostrum supply in the preceding phase of life, and concentration of inflammatory and stress parameters in blood and saliva in the rearing phase. The aim of the study is to clarify whether improved intestinal health through feeding prebiotics has a preventive effect against tail biting and whether the sum of all negative influencing factors including stress experienced since birth, which may be reflected in laboratory diagnostic parameters (acute phase proteins, stress markers), lead to the occurrence of tail biting after a so far undefined threshold has been exceeded.

The population of the native grey seals (Halichoerus grypus) developed well after their protection and subsequent recolonisation of the Wadden Sea. The positive trend of the population poses a challenge from the management perspective. There is an increasing requirement of suitable habitat, which is commonly associated with enhanced conflict potential. A special kind of conflict exists on the Heligoland Dune, where the largest grey seal colony occurs in German waters. Individual animals habituated to human presence due to spatial density and the continuous close contact between humans and animals and therefore do not display the expected escape reflex anymore. This involves an increased risk potential for swimmers, especially in the water. This study aims to test specific management measures in order to create a swimming area safe for people and to implement them officially if successful. On that account, an acoustic method shall be tested, which has not been used for that purpose yet and is based on the so called startle reflex. In contrast to conventional acoustic devices scaring off seals unspecifically, a very specific signal will be used, which has got no extensive negative effects on target and also non-target species in theory. The study investigates if the application of this system has got the desired efficiency and causes no evident negative effects in this special context.

Results:

Within the framework of the project work already carried out, it has not yet been possible to achieve the targeted goals, as unexpected problems have arisen during data collection in the field. Considering the importance of this system for the community of Helgoland, as well as for the acceptance of grey seals by the inhabitants and guests of Helgoland, it would be important to complete this started experiment. Through a desired project extension, an objectively evaluable data basis should be created, wherewith the acoustic deterrence system can be evaluated.

Vom zentralen Nervensystem absteigende inhibitorische Schmerzbahnen sind ein Teil des körpereigenen Systems zur Schmerzmodulation. Wird dieses System stimuliert, werden nachfolgende, schmerzhafte Reize als weniger schmerzhaft wahrgenommen, im englischen Sprachgebrauch wird das als "Diffuse noxious inhibitory control (DNIC)" bezeichnet. In der humanmedizinischen Forschung wird zur Evaluation der DNIC ein standardisierter, konditionierender Stimulus (CPM= conditioned pain modulation) eingesetzt, bevor der eigentliche Teststimulus erfolgt. Ist die DNIC intakt, wird der Teststimulus nach CPM als weniger schmerzhaft als vor/ ohne CPM wahrgenommen.
Für Labornager und in der Humanmedizin konnte gezeigt werden, dass die DNIC bei Individuen mit chronischen Schmerzen weniger stark ausgeprägt eiterhin ist für die Humanmedizin beschrieben, dass Patienten, die vor einer Operation niedrigere Werte im DNIC-Test aufweisen ein erhöhtes Risiko besitzen postoperativ an chronischen Schmerzen zu leiden
Für den Hund ist eine Technik zur Evaluierung der DNIC beschrieben. Eine systematische Evaluierung dieser Testmethodik, bezüglich Zuverlässigkeit und Reproduzierbarkeit beim Hund wurde bisher allerdings nicht durchgeführt.
Die Überprüfung der DNIC kann zu einer besseren Typisierung des vorliegenden Schmerzphenotypes und damit zu einer besseren Anpassung einer effektiven Schmerztherapie beitragen.

Im Rahmen dieses Forschungsprojektes soll eine standardisierte Methode zur Untersuchung der absteigenden inhibitorischen Schmerzbahnen beim Hund erarbeitet und kritisch evaluiert werden.
