In diesem Projekt werden in einem transgenen Mausmodell die Folgen einer Sertoli Zell-spezifischen Deletion des Connexin45-Gens auf die Regulation der Spermatogenese u.a. mittels IHC und WB untersucht. Die zelluläre Lokalisation von Connexin45 im Hoden adulter Mäuse soll mittels Cx45EGFP-Mäusen bestätigt werden.

Project Details:
Die testikuläre intraepitheliale Neoplasie (TIN) ist die Vorläuferläsion testikulärer Keimzelltumore, welche die häufigsten malignen Neoplasien bei jungen Männern darstellen, mit steigender Inzidenz. Ziel dieser Studie ist, Erkenntnisse zur Pathogenese zu gewinnen, die aufgrund der bereits intrauterinen Entstehung der TIN und ihrer pubertären Progression zum invasiven Tumor sowie bislang fehlender Zellkultur- und Tiermodelle nicht geklärt ist. Offen ist insbesondere die Rolle der Sertoli Zellen, welche in ihrer Differenzierung und Funktion in TIN-Tubuli verändert sind. Hierbei soll der Frage nachgegangen werden, ob die veränderten Sertoli Zellen eine Ursache der Fehlentwicklung der Keimzellen darstellen oder ob sich die Sertoli Zellen sekundär unter dem Einfluss der TIN verändern. Anhand einer Ko-Kultur adulter humaner Sertoli Zellen mit Seminomzellen sollen der gegenseitige Einfluss der Zellen auf Differenzierung, Proliferation und Funktion untersucht sowie mögliche Signalmoleküle identifiziert werden. Dazu werden Differenzierungsmarker, Zell-Zell-Kontakte, morphologische Merkmale sowie Proliferationsraten bestimmt und funktionelle Untersuchungen zu Zell-Zell-Kontakten durchgeführt. Die Ergebnisse werden mit denen humaner Hodenbiopsien verglichen. Die Studie soll zur Etablierung eines Zellkulturmodells zur Pathogenese testikulärer Keimzelltumore führen, die Interaktion zwischen Tumor- und Sertoli Zellen beleuchten und somit zum Verständnis der Entstehung und Entwicklung dieser Tumore beitragen.

Project Details:
Nach einer Ölpest kontaminieren Öl verschiedener Qualitäten und Ölgemische das Gefieder von Seevögeln. Nach Hautkontakt werden unter anderem auch Intoxikationen via transdermaler Permeation vermutet. Untersuchungen an Haut verschiedener Vogelarten in Franz-Zellen sollen die Qualität solcher Vergiftungen in Hinblick auf die Triage bewerten.

Project Details:
In diesem transgenen Tiermodell werden die Folgen einer Deletion von Connexin43 in Keimzellen auf die Entwicklung des Hodens und die Spermatogenese histologisch und immunhistochemisch untersucht. Verwendet werden hier Keimzell-spezifische VASA-Cre und Cx43 gefloxte Mauslinien.

Project Details:
Mit Hilfe verschiedener molekularbiologischer (z.B. Microarray Analysen, RT-PCR, qRT-PCR) und zellbiologischer (WB, IHC, IF) Methoden werden in diesem Projekt präpubertäre Mäuse (Tag 8 p.p.) der verschiedenen Genotypen des SCCx43KO-Mausmodells von Tag 8 untersucht. Interessante Gene (z.B. DMRT6, DMRT7, SOHLH1, etc.) werden vergleichend an humanen Hodenbiopsien mit normaler und gestörter Spermatogenese untersucht.

Project Details:
Erkrankungen der Beckenregion des Pferdes stellen u.a. aufgrund der starken Bemuskelung dieser Region und der Unzugänglichkeit einzelner Strukturen oft eine diagnostische Herausforderung dar. Zum Teil ist die Wertigkeit von klinischen und bildgebenden Befunden im Beckenbereich hinsichtlich ihrer Bedeutung als Lahmheitsursache unklar.
Pferde mit Erkrankungen der Beckenregion (d.h. des knöchernen Beckens, der Beckengürtelmuskulatur, des Hüftgelenks, des Kreuz-Darmbein-Gelenks, des Lumbosakral-Gelenks) werden klinisch, ggf. mittels diagnostischer Betäubungen, ultrasonographisch und röntgenologisch untersucht. Es erfolgt eine vergleichende Auswertung der bildgebenden Befunde hinsichtlich ihres diagnostischen Wertes, ggf. unter Einbeziehung von Sektionsbefunden. Die Erkrankungen werden falls möglich medikamentell oder anderweitig behandelt, und der Behandlungserfolg wird langfristig nachverfolgt.

Project Details:
A central question in evolutionary and conservation biology is how and to what extent species adapt to habitat fluctuations in their natural environment and to what extent environmental flexibility is linked to distribution and speciation. We address these questions in a longterm research programme by a comparative and integrative approach to nocturnal lemurs, a unique model for basal primates. Lemurs are endemic to Madagascar. Current research revealed a much higher species diversity in nocturnal lemurs than previously assumed. Lemurs underwent a broad-scale adaptive radiation after their initial colonization of Madagascar about 60 Mya. Extant lemurs consist of more than 100 species which inhabit the various forest habitats of the Island. Many species, in particular nocturnal species, have only been described recently, partly by our group. The underlying colonisation history and the factors driving this diversification are so far only poorly known. Here, we aim to understand the factors that promoted this radiation and shaped distribution, adaptation and genetic structures of lemurs. We are also addressing the question of the basis of reproductive isolation of species. These analyses are focused on the genera Microcebus spp., Lepilemur spp. and Avahi ssp. Whereas some species within a taxon show quite limited, others show broad distributions. Nocturnal lemurs belong to the endangered primates due to anthopogenically caused habitat degradation and poaching. We perform field studies, supplemented by experimental studies, particulary in north-western, northern and eastern Madagascar, using comparable methods, to study the ecology, behaviour, communication, parasite infections and the genetics of populations of lemurs of the same taxon showing contrasting distribution and to assess sensory, vocal, ecological and social determinants of species flexibility and its link to genetics. Our current research focus is on mouse lemurs, sportive lemurs and woolly lemurs. Results will deliver important tools to monitor and manage the biodiversity in tropical forests. Furthermore they will be a first step to understand the evolution of the unique species diversity of lemurs in Madagascar and the origin and evolution of our own primate ancestry.

Project Details:
There is an increasing concern on the effects of underwater noise on hearing of cetaceans. However, cochlear frequency maps (i.e. distribution of frequencies along the cochlear spiral within the inner ear) are still lacking for marine mammal species. The aim of this study is to predict the cochlear frequency map for harbour porpoise (Phocoena phocoena), based on morphometrics of the sensory cells of the organ of Corti (hearing organ). Once the map for a species is known, it will be possible to identify the frequencies that are impaired once lesions are found in the inner ear, and have better understanding of the consequences for individual to have a hearing loss in a particular hearing range. In cases of noise-induced hearing loss, it will be possible to extrapolate the characteristics of potential sound sources that have triggered a damage. This map will be a crucial tool for management of the effects of underwater noise on hearing in porpoises, as well as improved decision-making in conservation plans for harbour porpoises and other marine mammals.

In this project, a diagnostic examination of nutria shot in 2020 and 2021is carried out, and liver tissue is made available for chemical analysis. For this purpose, 41 individuals from Lower Saxony will be autopsied, including a histologic examination of lung, liver, kidney and spleen tissue. In addition, an individual age determination (adult animal/juvenile animal), sex, nutritional status, general health status and a sampling of liver tissue will be provided. Chemical residue analyses will be carried out by the BFG. The diagnostic findings are presented and summarized in an Excel sheet and final reports per sampling.

Results:

All animals showed typical traumatic, death-related injuries with histologically recognizable agonal changes in the lungs and the circulatory system. In addition, low-grade diffuse liver lipidoses were found in three animals, and high-grade urolithiasis was found in one animal. Five animals were pregnant females. Chronic interstitial pneumonia with a suspected infectious, presumably viral cause was diagnosed in 27 animals.
