Es wird ein effizientes, kostengünstiges Testsystem etabliert, um eine hohe Anzahl von Personen möglichst schnell
(> 1.000/Tag) zu screenen. In drei Prozessschritten werden i) Proben in mobilen Testzentren entnommen
(Rachenabstrich nach Standard des Robert-Koch-Instituts), ii) aufgearbeitet (RNA-Isolation) und iii) mittels PCRAnalytik nach dem Protokoll der Charité Berlin1 ausgewertet. Positive Ergebnisse aus dem Screening werden
diagnostisch über zertifizierte Labore bestätigt und dem lokalen Gesundheitsamt gemeldet. Die mobilen Stationen
werden auf Basis von flexibel aufstellbaren Schiffscontainern aufgebaut und sowohl für die Abstrichentnahme
als auch die Aufreinigung eingerichtet. Auf diese Weise wird eine strikte Trennung zwischen der Entnahme und
der Aufarbeitung potentiell infektiösen Materials im mobilen Container und der anschließenden Analytik der
nicht-infektiösen RNA-Proben in separaten Laboren sichergestellt.

diese in den meisten Geweben in vivo deutlich niedriger sind. Im Falle einer Infektion und der daraus resultierenden Entzündung kommt es zu einer massiven Absenkung der Sauerstofflevel, zu einer sogenannten Hypoxie, die die Wirt-Pathogen-Interaktion wesentlich beeinflussen kann. Zahlreiche Studien inklusive aktueller Publikationen der Antragsteller belegen, dass die Sauerstoffverfügbarkeit signifikante Auswirkungen auf zelluläre Prozesse im Laufe einer Infektion hat. Untersuchungen, welche die Wirts-Erregerabwehr unter physiologischen Sauerstoffbedingungen charakterisieren, sind daher unbedingt von Nöten, um die grundlegenden Reaktionen der Zelle besser zu verstehen und neue therapeutische und prophylaktische Ansätze entwickeln zu können. Leider gibt es bisher unzureichende Daten darüber, wie stark die Sauerstoffwerte im infizierten Gewebe tatsächlich absinken, um diese Bedingungen dann auch für molekulare und zelluläre in vitro Versuche umsetzen zu können.
Das Ziel dieser Studie ist es, die physiologisch und pathophysiologisch relevanten Sauerstofflevel im Laufe einer bakteriellen zoonotischen Infektion zu detektieren, um diese für in vivo nahe Studien der Wirt-Pathogen-Interaktion anwenden zu können. Im Fokus des hiermit beantragten Versuchsvorhabens steht als Beispiel die Erforschung der Interaktion von Streptococcus (S.) suis als zoonotischer Krankheits- und Modellkeim mit dem Schwein als natürlichen Wirt sowie das Modell für die S. suis Meningitis beim Menschen. Da die Sauerstoffbedingungen während einer bakteriellen Meningitis wie auch bei anderen Infektionsgeschehen im Vergleich zum gesunden Tier im Liquor nicht bekannt sind, ist zunächst das erste Ziel, die Sauerstofflevel während in vivo Tierversuchen genau zu charakterisieren. Zusätzlich sollen die Systeme für die Sauerstoffmessungen auch für weitere Gewebetypen in vivo angepasst und etabliert werden, um die technische Expertise zukünftig Kooperationspartnern zur Verfügung stellen zu können. Schließlich sollen im Labor etablierte porcine und humane in vitro Systeme an die detektierten in vivo physiologischen und pathophysiologischen Sauerstofflevel für Studien der Wirt-Pathogen-Interaktion sowie der Suche nach neuen therapeutischen Ansätzen angepasst werden.
Die zu erwartenden Ergebnisse stellen sowohl technisch wie auch inhaltlich einen hohen Mehrwert für die vernetzte Zoonoseforschung dar und sollen mittels eines Workshops an Mitglieder der Zoonoseplattform weitergegeben werden. Die Zusammenarbeit der Zoonosenplattform mit den Antragstellern soll dazu dienen, technische Expertise, Erfahrungen und Wissen auszutauschen. Dadurch sollen zukünftig neue innovative Projekte mit weiteren Erregern über die Speziesgrenzen in Kooperation mit Mitgliedern der Zoonoseplattform basierend auf der Thematik "Hypoxie" und den im Rahmen dieses Projektes etablierten Techniken imitiert werden.

Streptococcus (S.) suis is one of the most important pathogens in pigs and also an emerging zoonotic agent, causing meningitis and various other pathologies. The pathogenesis of S. suis infections is highly complex and still only poorly understood. Infiltrations with high numbers of neutrophils are typical for S. suis diseases. Neutrophil extracellular traps (NETs) are an important defence mechanism against different pathogens, but its role during S. suis infections and also during meningitis in general, has not been studied so far. Therefore, the goal of the proposed project is to explore the role of NETs in the pathogenesis of S. suis meningitis focussed on the cerebrospinal fluid compartment. Preliminary experiments revealed that S. suis can induce NET-formation in porcine and human blood-derived neutrophils. On the other hand, S. suis also exhibited the ability to evade entrapment by NETs using the DNases SsnA and EndAsuis to degrade NETs. Thus, this project is designed to investigate the role of NETs in the pathogenesis of S. suis meningitis. Human and porcine models of the blood-cerebrospinal fluid barrier as well as in vivo studies using the pig as natural host will be used to elucidate the impact of NET-formation and NET degradation on the host cell-pathogen interaction in the cerebrospinal fluid compartment. The cooperation of both partners, C. Baums (Leipzig) and M. von Köckritz-Blickwede, with their expertise in S. suis pathogenesis and NET-formation, respectively, provides an excellent condition for the realization of this project.

Der Transkriptionsfaktor HIF-1alpha ist ein Hauptregulator der Homöostase und zellulären Anpassung an Sauerstoffstress. Es gibt zunehmend Hinweise dafür, dass HIF-1alpha durch bakterielle Pathogene aktiviert werden kann und die bakterizide Aktivität von Phagozyten beeinflusst. HIF-1alpha-Agonisten, die in der Lage sind, antibakterielle Mechanismen der Wirtsimmunabwehr zu verstärken, könnten neben Antibiotika zur unterstützenden Immuntherapie insbesondere gegen Antibiotika-resistente Keime wie methicillin-resistente Staphylococcus aureus (MRSA) Anwendung finden. Allerdings sind die detaillierten Mechanismen, wie HIF-1alpha die Immunzellen beeinflusst, immer noch unzureichend geklärt. Eigene Vorarbeiten zeigen, dass HIF-1alpha-boosting die Bildung von extrazellulären DNA-Netzen induziert. Diese sogenannten phagocyte extracellular traps (PETs) wurden kürzlich als neuer wichtiger Immunabwehrmechanismus gegen Infektionen bekannt. Das Ziel dieser Studie ist es (i) Zellen zu identifizieren, die PETs in Abhängigkeit von HIF-1alpha
während der Infektion oder unter hypoxischen Bedingungen ausschleusen, (ii) die Mechanismen der HIF-1alpha-abhängigen PET-Bildung zu charakterisieren und (iii) die Rolle der IF-1alpha-abhängigen PET-Bildung in vivo gegen MRSA-Infektionen zu untersuchen. Diese neuen wissenschaftlichen Erkenntnisse
sollen wichtige Grundlagen für eine HIF-1alpha-abhängige Immunantwort als neuen therapeutischen Ansatz liefern.

Infectious diseases are a worldwide problem, sometimes causing declines and extinctions in wild populations and species. An alarming situation is the infestation of amphibians by a chytrid fungus: Batrachochytrium dendrobatidis (Bd) has infected over 520 species worldwide. This fungus grows in the skin surface of amphibians and keratinizes cells; this difficults the transport of ions, and therefore osmoregulation, which in many cases leads to death by cardiac arrest and massive mortalities. Habitat fragmentation, within other factors including diseases, has lead amphibians to be the most threatened vertebrate group. In Europe, the yellow bellied toad (Bombina variegata) is classified as endangered, and is one of the few anurans with a high Bd infection prevalence. In Germany, yellow bellied toads have been recorded to have a Bd infection rate around 14%, however no Bd massive mortalities due to chytridiomycosis have been reported. In spite of that, Bd infection could entail a fitness reduction. Currently the yellow bellied toad is listed at the German red list as endangered and within some regions of Germany as threatened with extinction. For this reason the NABU started a native habitat management and connection project, which includes reintroductions. As Bd is present in Germany, this project aims to understand better the threat Bd is posing to yellow bellied toad populations. At the northernmost distribution of the yellow bellied toad, NABU is monitoring 16 populations and in four of them toads have been reintroduced. These populations were sampled three times in spring and summer 2016 (May, July and September) to determine the difference in infection rate in relation to temperature, population size and genetic diversity. The quantification of Bd infection rate is determined by real-time qPCR. The technique was established at RIZ-TiHo by Dr. Bourke together with de Buhr, PhD. Up to now, 593 DNA samples have been extracted, the qPCR technique set up and Bd infection quantified.

Durch die Entwicklung der antimikrobiellen Therapie konnte die Morbidität und Mortalität von Infektionskrankheiten erheblich reduziert werden. Allerdings hat die Entstehung von resistenten Mikroorganismen jetzt epidemische Ausmaße erreicht und stellt große Herausforderungen an die Human- und Veterinärmedizin. Für jedes derzeit verfügbare Antibiotikum haben potentielle Pathogene Resistenzen entwickelt. Beunruhigend ist dabei, dass in mehreren wichtigen Gram-positiven Bakterienarten wie Staphylococcus (S.) aureus, die Resistenzen zu einem Therapieversagen bei der Verwendung von Standardantibiotika führt. Neue Stämme des Methicillin-resistenten S. aureus (MRSA) verursachen schwere zoonotische Infektionen und können vom Tier auf den Menschen oder umgekehrt übertragen werden. Weiterhin sind subklinische S. aureus Mastitiden sehr häufig ein Problem in der Praxis. Innovative Ansätze zur Behandlung werden dringend benötigt. Ein alternativer Ansatz für die Behandlung von schweren Infektionen, ist die pharmakologische Verstärkung der antimikrobiellen Fähigkeiten von phagozytischen Immunzellen und eine dadurch resultierende Stärkung des Wirtsimmunsystems. Das Ziel dieser Studie ist es zu erforschen welche Wirkung bovine Cathelicidine gegen bovine bakterielle Mastitiserreger haben. Dabei soll zum einen untersucht werden, über welche antimikrobielle Wirkung bovine Cathelicidine selbst verfügen und inwieweit sie Phagozyten in ihrer Immunabwehr stärken. Letztlich sollen in einem Zellkultursystem der Blut-Euterschranke Wirkungen für spätere Applikationen erforscht werden.

Until now only little is known about the impact of physiological oxygen conditions (physioxia) on the viability, differentiation process, and the antimicrobial activity of immune cells like mast cells (MCs). MCs are important innate immune cells mediating immune cell recruitment and antimicrobial defense. They originate from hematopoietic precursors and specifically differentiate to mature and functional MCs in the destination tissue such as skin, mucosa and intestine. To differentiate MCs and to study their immune cell function, atmospheric oxygen conditions (20-21% O2) are traditionally used in cell culture although the physiological oxygen conditions in vivo in tissues are significant lower. This study aims to characterize the differentiation of immature murine bone marrow-derived MCs under physiological oxygen conditions (7% O2; 53 mmHg). Therefore, bone marrow derived suspension cells were cultured in the presence of IL-3 for 39 days with continuous, non-invasive determination of the oxygen level using a Fibox4-PSt3 measurement system without technique-dependent oxygen consumption. Cellular viability and differentiation status were monitored with cell specific surface marker using flow cytometry. The impact of physioxia on the maturation of MCs was ascertained by investigating histamine level with high performance liquid chromatography and quantifying the expression of the transcription factor hypoxia-inducible factor 1 (hif-1á) and its target genes vegf, il-6, and tnf-á with real time PCR. Using toluidine blue staining we confirmed that the differentiated MCs stay viable under physioxia during the specified period of 39 days. Interestingly, MCs cultivated at 7% O2 (53 mmHg) show a significantly delayed differentiation rate defined by CD117-positive cells compared to MCs cultivated under normoxia (21% O2). Investigating the gene expression of hif-1á and its target genes of differentiated MCs exhibited a significant lower transcript expression under physioxia compared to normoxia.
