Mouse lemurs are the smallest primates on earth. They are endemic to Madagascar, where they are increasingly endangered by anthropogenically caused habitat degradation. Mouse lemurs are unique primate models for evolutionary, aging and genomic research. They exhibit extraordinary species diversity with limited vs. broad ranges in the Malagasy tropical forests. They have a long longevity related to body size (up to 15 years in the laboratory, about 9 years in the field) which is, however, much shorter than for common anthropoid primate models. Furthermore, some, but not all, aging individuals develop an AD-like pathology as found in Alzheimer patients or show human-like aging-related diseases such as cancer or ocular pathologies (e.g. cataracts). Likewise mouse lemurs are one of the primate models, for which the full genome is published. In 1985, Elke Zimmermann founded the colony of gray mouse lemurs of the Institute of Zoology (IfZ), by two founder pairs coming from the University of Tübingen (Jörg Ganzhorn). Founder pairs stem from the Rotterdam Zoo and were tracked back to their origin in south-eastern Madagascar (Mandena). To increase the genetic diversity of the colony, the offspring of these founders (originally based at the University of Stuttgart-Hohenheim, moving later on to the University of Constance, the German Primate Centre and finally completely to the Institute of Zoology at the University of Veterinary Medicine in Hannover) were cross-bred with mouse lemurs coming from the Rotterdam Zoo, the Netherlands, in 1993 and the Parc Zoologique de Vincennes, France, in 1996. The colony is a self-sustaining breeding colony for which the life history of each individual mouse lemur is documented from birth to death in a mouse lemur database. Tissues from these individually known mouse lemurs are collected for veterinary purposes, or after the natural death of an animal. This colony of mouse lemurs is registered in the European and International Studbook of the Cheirogaleidae. Animals are used for non-invasive integrative and comparative research in the field of behaviour, physiology, reproduction, sensory biology, communication, cognition, genetics/genomics, aging, conservation biology and veterinary medicine. Furthermore, they are used for comparative research projects that are conducted together with several academic institutions studying the effect of different factors (e.g. general maintenance conditions, different photoperiod regimes, enrichment, group composition, genetic lineages) on the physiology, behaviour, communication, cognition, reproduction, longevity and health status of the colonies.

Die Erfassung und Bewertung von Emotionen bei Tieren gewinnt zunehmend an Interesse. Zum einen weil die Öffentlichkeit zunehmend für den Tierschutz sensibilisiert ist zum anderen da Labortiere als Modellspezies für die Entwicklung von Psychopharmaka dienen. Die zuverlässige Bewertung von Emotionen ist nach wie vor eine wissenschaftliche Herausforderung, da Emotionen nicht nur das Ergebnis automatischer physiologischer Reaktionen sind, sondern auch einer kognitiven Bewertung unterliegen (Kalat & Shiota, 2007). So kann die emotionale Wahrnehmung einer Situation von individuellen Faktoren wie der eigenen Persönlichkeit oder Erfahrung abhängen. Um Tierwohl bewerten zu können, ist es von Bedeutung verlässliche Methoden zur Erfassung des emotionalen Zustandes eines Tieres zu entwickeln. Zwar gibt es Möglichkeiten den Zustand eines Tieres über physiologische Messungen (z.B. Blutabnahme, Implantate zur Messung der Herzfrequenz) zu erfahren, jedoch sind diese Maßnahmen invasiv und geben keine Aussage über die emotionale Valenz des Zustandes (eine erhöhte Herzfrequenz tritt sowohl bei negativen als auch bei positiven Emotionen auf). Eine Alternative bieten Verhaltenstests mit denen Persönlichkeit von Tieren als auch die emotionale kognitive Bewertung einer Situation getestet werden kann. Eine Möglichkeit die kognitive Bewertung zu testen, ist die "Judgement Bias Task"". Die Tiere lernen einen positiven und einen weniger positiven Stimulus zu unterscheiden und entsprechend zu reagieren. Anschließend wird ihnen ein unbekannter ambivalenter Stimulus präsentiert. An ihrer Reaktion lässt sich einschätzen, ob ein Tier ""optimistisch"" (wählt wie bei positivem Stimulus) oder ""pessimistisch"" (wählt wie beim weniger positives Stimulus) ist. Diese Erkenntnisse werden eine bessere Bewertung von Emotionen bei Tieren erlauben und können im späteren Verlauf genutzt werden, die Auswirkung verschiedener Haltungsbedingungen, Umgebungsanreicherungen oder pharmakologischer Effekte auf die Emotionen eines Tieres zu untersuchen."

This project serves the long-term purpose of protecting the species of the yellow-bellied toad, Bombina variegata, which is highly endangered in Germany and Central Europe. In many areas, the yellow-bellied toad only occurs in secondary, i.e. anthropogenically modified, habitats such as quarries. The populations are often isolated, too small, and suffer from inbreeding and genetic impoverishment. Previous studies have also shown that yellow-bellied toads use different life cycle strategies in different habitats. These differences suggest that the species has adapted to local ecological conditions, and these adaptations are potentially manifested in the genome. The project aims to improve the genetic and ecological information available about the species in order to make practical conservation more effective. For this purpose, DNA samples from German, French and Swiss yellow-bellied toad populations, which have already been sampled in previous projects, are examined using genomic methods (RADSeq). In addition, toads in Germany will be sampled in the few natural habitats where they still occur. The genomic results are then correlated with ecological data (habitat type, local climate). The results provide evidence as to whether yellow-bellied toads are adapted to their local habitat (anthropogenic vs. natural) or to their local climate at the molecular level. The aim is to identify specific candidate genes that mediate this local adaptation. Furthermore, species distribution models based on climatic data should show where suitable habitats are for the yellow-bellied toad today and in the future - also with a view on climate change. The results obtained will be discussed with scientists and species protection specialists in a workshop. In follow-up projects, so-called genetic rescue will be used for small, isolated occurrences to stabilize them, and larger populations will be genetically upgraded. Genetic rescue projects based on genomic information are innovative since so far they have been successfully implemented in species protection in only a few countries.

Aposematism and crypsis are contrasting antipredator strategies. In the color polymorphic Oophaga pumilio, phylogenetic analyses have shown that populations with cryptic and aposematic coloration might have evolved independently in different islands. Two other species, O. granulifera and O. vicentei also display cryptic and aposematic color morphs in geographically isolated populations. Particularly, phylogenetic analyses of the evolution of these contrasting antipredator strategies can improve our understanding on the evolution of polymorphism in aposematic species. This project aims to uncover the molecular signatures of selection present in the genes of three Oophaga species that experienced independently evolved cryptic and aposematic phenotypes. To this effect, we look forward to elucidate the phylogenetic relationships of the genus and quantify the variation in gene expression and allelic frequencies using state-of-the-art methods.

Mit Sprachassistenten wie Alexa, Siri und Cortana hat die Spracherkennungs- und Steuerungssoftware einen festen Platz in unserem Alltag gefunden. Auch Tierstimmen enthalten eine Fülle an Informationen über das Sendertier, wie beispielsweise Art, Identität, Geschlecht, Alter, Körpergröße, emotionalen Zustand, Krankheiten und Hormonstatus. Die Bioakustik ermöglicht somit die Entwicklung von non-invasiven, zeit- und kosteneffizienten, automatischen bioakustischen Überwachungssystemen von Tieren. Hierfür werden Mikrofone installiert, die die Lautäußerungen der Tiere rund um die Uhr aufzeichnen, ohne sie zu stören oder zu beeinflussen. Die Aufnahmen können mithilfe mathematischer Algorithmen automatisch analysiert werden, um spezifische Informationen zu extrahieren. Darüber hinaus können psychoakustische Werkzeuge eingesetzt werden, um gezielte positive Verhaltensweisen hervorzurufen. Solche bioakustischen Werkzeuge können sowohl zur Klarifizierung taxonomischer Fragen, Überwachung von Wildtieren als auch zur Verbesserung von Tierhaltungen eingesetzt werden und spielen somit zunehmend eine wichtige Rolle im Artenschutz und im Tiermanagement. Das Ziel dieses Projekts ist es, die vielfältigen Anwendungsmöglichkeiten zu erkunden und bioakustische Monitoring Systeme für Artenschutz und Tiermanagement zu entwickeln.

Oophaga pumilio, besser bekannt als Erdbeerfrosch, gehört zur Familie der Pfeilgiftfrösche (Dendrobatidae). Diese winzigen, tropischen Frösche fallen sofort durch ihre intensiven Farben auf. Innerhalb der Art O. pumilio besteht ein ausgeprägter Farbpolymorphismus, der sich in einer Vielzahl von Hautfarben und Mustern äußert. Kryptische Individuen zeigen meist eine grüne, unauffällige Färbung, während aposematische Vertreter durch leuchtende Farben von intensivem Rot bis zu strahlendem Gelb charakterisiert sind. Die Farbintensität von O. pumilio spielt somit eine zentrale Rolle sowohl bei der Prädatoren Abwehr, weitere Studien haben gezeigt, dass die unterschiedlichen Farben eine Rolle bei der Partnerwahl spielen. Zudem ist bekannt, dass die Farbrezeptoren in der Retina den Fröschen ermöglichen zwischen den Farbmorphen zu unterscheiden und Carotenoidpigmente für die Ausprägung der Färbung eine zentrale Rolle spielen. Carotinoide sind fettlösliche Pigmente, die für rosafarbene, rote, orange und gelbe Färbungen verantwortlich sind. Da O. pumilio keine Carotinoide selbst herstellen kann, muss die Aufnahme von Carotinoiden über tierische Beutetiere erfolgen. Nach der Aufnahme werden die Carotinoide im Gastrointestinaltrakt resorbiert, metabolisiert und anschließend in der Haut sowie in weiteren Organen, etwa den Augen, gespeichert. Im Rahmen eines Ernährungsversuchs sollen die Frösche mit Carotinoiden supplementiert werden, um den Einfluss dieser Pigmente auf die Hautfärbung zu untersuchen und ein besseres Verständnis der Farbentwicklung bei O. pumilio zu gewinnen.
Das natürliche Verbreitungsgebiet von O. pumilio umfasst Westpanama, die Karibikküste Costa Ricas sowie Teile Nicaraguas. Die Art bewohnt tropische Regenwälder mit dichter Vegetation und wird häufig an Waldrändern in Küstennähe beobachtet. Angesichts ihres durchschnittlichen Körpergewichts von rund einem halben Gramm stellt sich weiterhin die Frage, wie sich diese kleinen Tiere in den komplexen Strukturen dichter Regenwälder in ihren Territorien von mehreren Quadratmetern orientieren. Zur Beantwortung dieser Fragestellung werden zwei unabhängige Verhaltensversuche durchgeführt, die das räumliche Lernverhalten anhand von visuellen Signalen, die sich in ihren Farben unterscheiden, von Oophaga pumilio untersuchen sollen.

The overall objective of this project is to understand the effects of forest fires on vegetation and wildlife dynamics in dry deciduous forests of western Madagascar and to model their long-term post-fire regeneration potential by applying a landscape approach. This study will build on the fire history of the last 35 years within Ankarafantsika National Park (ANP) derived from annually available remote sensing data. We aim to investigate the impact of different time spans since the last fire, fire severity and different fire intervals on forest integrity on the one hand, and on lemur population dynamics as one representative and enigmatic, forest-dependent, vertebrate group on the other hand. Lemurs are highly suitable study species in this respect, since they fulfill key ecosystem functions (e.g. pollination, seed dispersal, food-web interactions), and the presence of the complete lemur assemblage indicates intact forest habitats. This will allow a prognosis of the long-term regeneration potential of recently burnt areas. As such, this project will provide guidance for a sustainable conservation management in the region and beyond, and will help to identify preferable areas for forest restoration and therefore nature-based-solutions towards pressing environmental problems in Western Madagascar.

Aposematismus ist die Assoziation zwischen einer auffälligen Warnfärbung und einem Abwehrmechanismus (z. B. Gifte) zur Vermeidung von Prädation. Die Genetik der Färbung und der Giftigkeit bei aposematischen Tieren ist noch nicht gut erforscht. In diesem Zusammenhang ist die Vielfalt der Farbmuster und der Alkaloide in der Haut, die mit Strategien zur Vermeidung von Raubtieren bei neotropischen Giftfröschen verbunden sind, eine exzellente Gelegenheit, die Konvergenz der ökologischen Anpassung zu untersuchen. Pfeilgiftfrösche unterscheiden sich in ihren Farbmustern und ihrer Toxizität zwischen den Populationen und es gibt starke Beweise dafür, dass Aposematismus adaptiv ist, wahrscheinlich angetrieben durch die Selektion von Raubtieren. Für den Erdbeerfrosch Oophaga pumilio konnten wir für Gene, die im Zusammenhang mit dem Pigment-Metabolismus stehen (z.B. Pteridingene), zeigen, dass sie zwischen Farbmorphen unterschiedlich stark exprimiert sind. Nächstes Ziel ist es, den Aufbau pigmenthaltiger Zellen (Chromatophoren), die chromatographische Charakterisierung von Farbpigmenten und Alkaloiden, Expressionsmuster von Genen, die an der Färbung und Giftigkeit beteiligt sind, für drei farbpolymorphe, neotropischer Pfeilgiftfrösche der Gattung Oophaga in Verbindung zu setzen.
Für dieses Projekt werden wir Haut- und Leberproben von acht Populationen mit entweder grüner oder roter Färbung in Panama und Costa Rica sammeln. Zusammen mit Mitarbeitern der Universidad de Costa Rica, der TiHo, der University of Cambridge und des STRI in Panama werden wir die Chromatophoren in der Haut elektronenmikroskopisch charakterisieren, die Farbpigmente und Toxine in der Haut gaschromatographisch analysieren und eine RNAseq-Analyse mit dorsaler Haut durchführen. Wir erwarten, dass die Expressionsniveaus von Genen im Pigment- (z.B. Carotinoide) und Alkaloidmetabolismus zwischen verschiedenen Farbmorphen (rot vs. grün) variieren, aber aufgrund der Konvergenz innerhalb der Farbmorphen zwischen den Arten ähnlich sind. Unsere Studie wird zu einem besseren Verständnis des Zusammenhangs zwischen Geno- und Phänotypen adaptiver Anti-Räuber-Strategien führen und die Evolution regulatorischer Mechanismen der Genexpression beleuchten.

Drivers of speciation are diverse and complex, ranging from large-scale geomorphological processes like the formation of mountains or rivers to small-scale mechanisms like intra-specific ecological plasticity, divergent habitat choice or colonization potential. Due to the lack of appropriate model regions and species, previous studies typically focused on single or dual factor approaches without investigating interactions between drivers. We propose a highly suitable model region in eastern Madagascar to better explore the principal evolutionary processes driving species diversification and their interactions. The proposed study region is traversed by several large rivers that differ greatly in age. The geology and vegetation is rather uniform with similar macro-habitats ranging from tropical lowland to highland rain forest along an elevational east-west gradient. The area harbors an outstanding lemur species richness of which three genera are chosen as models for this project. Thirteen model species belong to two exceptionally speciose genera, Lepilemur and Microcebus, in contrast to the less speciose but ecologically plastic genus of woolly lemurs (Avahi). Rivers and mountains are assumed to have played a crucial role during speciation of lemurs, providing refugia during Pleistocene glaciation events which supposedly facilitated speciation. We propose that river chronology is an additional major co-variate shaping diversification processes, colonization pathways and phylogeographic history of species. We aim to combine geomorphological reconstructions of relative river ages, (micro-)habitat characterizations, ecological niche modeling techniques and RADseq-based phylogeographic reconstructions of colonization routes to investigate the role and interactions of various drivers of speciation including chronology of riverine barriers, vagility, altitudinal tolerance, body size, and ecological plasticity. This interdisciplinary project will thereby illuminate the relative importance of various extrinsic (e.g., geomorphology, habitat) and intrinsic factors (e.g., ecological plasticity, vagility) for the evolutionary diversification of animal populations.

Ziel des Projekts ist es, erstmals einen Beitrag zur Aufklärung der vokalen Flexibilität eines vokal aktiven und stammesgeschichtlich basalen Primaten während der Entwicklung zu leisten. Unsere bisherigen Forschungsergebnisse an der basalen Primatenart Mausmaki deuten auf eine unerwartet hohe vokale Flexibilität in der frühen Ontogenese, die in der weiteren Entwicklung abnimmt. Zu ihrer Untersuchung schlagen wir drei verschiedene, miteinander verzahnte Arbeitsprogramme vor. Die Präsenz und Rolle von Brabbeln (babbling) und vokalem Austausch (vocal-turn taking) während der Entwicklung sowie potentielle Determinanten, die zu entwicklungsbedingten Veränderungen in der vokalen Struktur führen, wollen wir durch standardisierte bioakustische und videografische verhaltensbiologische Untersuchungen an einem Datensatz von insgesamt 26 Kindern währen der Entwicklung und ihren 13 Müttern erfassen. Über Maschinenlernansätze, die auch selbst-organisierende neurale Netze einschließen, wollen wir die Dynamik der Veränderungen während der Entwicklung automatisiert quantifizieren und mögliche, zu Grunde liegende Regeln ableiten. Über Larnyxexperimente, verbunden mit histologischen und MikroCT-Untersuchungen des Vokaltrakts sowie über Hochgeschwindigkeitsvideoaufnahmen von Mund- und Zungenbewegungen während des Vokalisierens, wollen wir einen ersten Einblick in die Mechanik der Lautproduktion eines basalen Primaten erhalten und mögliche biomechanische Zwänge der vokalen Flexibilität erfassen. Die integrative Bearbeitung der verschiedenen Datensets wird eine Validierung der eingesetzten Methoden erlauben. Das Vorhaben stellt eine wichtige empirische Grundlage für die vergleichende akustische Kommunikationsforschung bei Säugetieren dar und trägt dazu bei, die stammesgeschichtlichen Wurzeln der vokalen Flexibilität in der Primatenkommunikation als wichtigen Baustein für die Evolution von Sprache zu verstehen.
