This project serves the long-term purpose of protecting the species of the yellow-bellied toad, Bombina variegata, which is highly endangered in Germany and Central Europe. In many areas, the yellow-bellied toad only occurs in secondary, i.e. anthropogenically modified, habitats such as quarries. The populations are often isolated, too small, and suffer from inbreeding and genetic impoverishment. Previous studies have also shown that yellow-bellied toads use different life cycle strategies in different habitats. These differences suggest that the species has adapted to local ecological conditions, and these adaptations are potentially manifested in the genome. The project aims to improve the genetic and ecological information available about the species in order to make practical conservation more effective. For this purpose, DNA samples from German, French and Swiss yellow-bellied toad populations, which have already been sampled in previous projects, are examined using genomic methods (RADSeq). In addition, toads in Germany will be sampled in the few natural habitats where they still occur. The genomic results are then correlated with ecological data (habitat type, local climate). The results provide evidence as to whether yellow-bellied toads are adapted to their local habitat (anthropogenic vs. natural) or to their local climate at the molecular level. The aim is to identify specific candidate genes that mediate this local adaptation. Furthermore, species distribution models based on climatic data should show where suitable habitats are for the yellow-bellied toad today and in the future - also with a view on climate change. The results obtained will be discussed with scientists and species protection specialists in a workshop. In follow-up projects, so-called genetic rescue will be used for small, isolated occurrences to stabilize them, and larger populations will be genetically upgraded. Genetic rescue projects based on genomic information are innovative since so far they have been successfully implemented in species protection in only a few countries.

Stable and consistent individual behavioral variations in animals that are generally regarded as an expression of animal personalities, and might be important drivers of ecological specialization and the evolutionary adaptive potential of species. We aim to investigate various proximate factors driving individual variations in risk-taking behavior of the grey mouse lemur (Microcebus murinus) as well as their relationship and explanatory value for physical cognitive performance. In the wild, this species lives under a very high predation pressure and risk taking decisions must therefore be taken on a daily basis and should be an important key to survival. We will study captive grey mouse lemurs with a longitudinal approach to investigate systematically the individual behavioral dynamics across time and different risk contexts. In a second step, personality differences will then be compared with the results from a test battery in the physical cognitive domain. Our findings will contribute to a better understanding of risk taking as one facet of animal personality in one of the world?s smallest primates.

Aposematism and crypsis are contrasting antipredator strategies. In the color polymorphic Oophaga pumilio, phylogenetic analyses have shown that populations with cryptic and aposematic coloration might have evolved independently in different islands. Two other species, O. granulifera and O. vicentei also display cryptic and aposematic color morphs in geographically isolated populations. Particularly, phylogenetic analyses of the evolution of these contrasting antipredator strategies can improve our understanding on the evolution of polymorphism in aposematic species. This project aims to uncover the molecular signatures of selection present in the genes of three Oophaga species that experienced independently evolved cryptic and aposematic phenotypes. To this effect, we look forward to elucidate the phylogenetic relationships of the genus and quantify the variation in gene expression and allelic frequencies using state-of-the-art methods.

Aposematismus ist die Assoziation zwischen einer auffälligen Warnfärbung und einem Abwehrmechanismus (z. B. Gifte) zur Vermeidung von Prädation. Die Genetik der Färbung und der Giftigkeit bei aposematischen Tieren ist noch nicht gut erforscht. In diesem Zusammenhang ist die Vielfalt der Farbmuster und der Alkaloide in der Haut, die mit Strategien zur Vermeidung von Raubtieren bei neotropischen Giftfröschen verbunden sind, eine exzellente Gelegenheit, die Konvergenz der ökologischen Anpassung zu untersuchen. Pfeilgiftfrösche unterscheiden sich in ihren Farbmustern und ihrer Toxizität zwischen den Populationen und es gibt starke Beweise dafür, dass Aposematismus adaptiv ist, wahrscheinlich angetrieben durch die Selektion von Raubtieren. Für den Erdbeerfrosch Oophaga pumilio konnten wir für Gene, die im Zusammenhang mit dem Pigment-Metabolismus stehen (z.B. Pteridingene), zeigen, dass sie zwischen Farbmorphen unterschiedlich stark exprimiert sind. Nächstes Ziel ist es, den Aufbau pigmenthaltiger Zellen (Chromatophoren), die chromatographische Charakterisierung von Farbpigmenten und Alkaloiden, Expressionsmuster von Genen, die an der Färbung und Giftigkeit beteiligt sind, für drei farbpolymorphe, neotropischer Pfeilgiftfrösche der Gattung Oophaga in Verbindung zu setzen.
Für dieses Projekt werden wir Haut- und Leberproben von acht Populationen mit entweder grüner oder roter Färbung in Panama und Costa Rica sammeln. Zusammen mit Mitarbeitern der Universidad de Costa Rica, der TiHo, der University of Cambridge und des STRI in Panama werden wir die Chromatophoren in der Haut elektronenmikroskopisch charakterisieren, die Farbpigmente und Toxine in der Haut gaschromatographisch analysieren und eine RNAseq-Analyse mit dorsaler Haut durchführen. Wir erwarten, dass die Expressionsniveaus von Genen im Pigment- (z.B. Carotinoide) und Alkaloidmetabolismus zwischen verschiedenen Farbmorphen (rot vs. grün) variieren, aber aufgrund der Konvergenz innerhalb der Farbmorphen zwischen den Arten ähnlich sind. Unsere Studie wird zu einem besseren Verständnis des Zusammenhangs zwischen Geno- und Phänotypen adaptiver Anti-Räuber-Strategien führen und die Evolution regulatorischer Mechanismen der Genexpression beleuchten.

The overall objective of this project is to understand the effects of forest fires on vegetation and wildlife dynamics in dry deciduous forests of western Madagascar and to model their long-term post-fire regeneration potential by applying a landscape approach. This study will build on the fire history of the last 35 years within Ankarafantsika National Park (ANP) derived from annually available remote sensing data. We aim to investigate the impact of different time spans since the last fire, fire severity and different fire intervals on forest integrity on the one hand, and on lemur population dynamics as one representative and enigmatic, forest-dependent, vertebrate group on the other hand. Lemurs are highly suitable study species in this respect, since they fulfill key ecosystem functions (e.g. pollination, seed dispersal, food-web interactions), and the presence of the complete lemur assemblage indicates intact forest habitats. This will allow a prognosis of the long-term regeneration potential of recently burnt areas. As such, this project will provide guidance for a sustainable conservation management in the region and beyond, and will help to identify preferable areas for forest restoration and therefore nature-based-solutions towards pressing environmental problems in Western Madagascar.

Ein vokales Repertoire enthält eine artspezifische Liste von Lautäußerungen, die von den Tieren produziert werden. Diese Listen bilden die methodische und theoretische Grundlage für eine Reihe von Forschungsfragen verschiedener Forschungsdisziplinen. Dennoch gibt es keine Einigkeit darüber, wie ein vokales Repertoire zu erstellen ist oder wie Ruftypen zu definieren sind. In den meisten Studien wurden die Ruftypen durch visuelle Klassifizierung anhand von Spektrogrammen bestimmt, die durch statistische Analysen auf der Grundlage der gemessenen akustischen Merkmale einer Vokalisation bestätigt wurden. In neueren Veröffentlichungen wurden objektivere mathematische Ansätze wie "supervised" oder "unsupervised" Clustering-Algorithmen entwickelt. Allerdings erfordern auch diese Algorithmen oft eine Vorklassifizierung des Beobachters, entweder, weil eine Anzahl von erwarteten Clustern bestimmt werden muss, oder, weil das Clustering-Ergebnis auf seine biologische Plausibilität hin überprüft werden muss. Beide Methoden können zu einer Überschätzung der Anzahl von Ruftypen führen, insbesondere bei Arten mit sehr variablen Vokalisationen oder bei Ruftypen mit graduellen Übergängen. Eine akustische Variation allein reicht also nicht aus, um eine Verhaltensrelevanz widerzuspiegeln. Dies erfordert auch eine Kontextspezifität der Rufe und die Fähigkeit des Gehirns, die akustische Variationen wahrzunehmen und verarbeiten zu können. In diesem Projekt erstellen wir vokal Repertoire, indem wir die mathematische Klassifizierung der Vokalisationen mit den Verhaltenskontexten, in denen die Rufe produziert wurden, sowie mit den physikalischen Hörfähigkeiten (z. B. Hörbereich, Zeit- und Frequenzauflösung) der jeweiligen Art kombinieren. Wir verwenden diesen Ansatz für zwei kleine Säugetierarten: die mongolische Wüstenrennmaus, ein wichtiges Tiermodell in der Hörforschung, und die Etruskerspitzmaus, die aufgrund ihres kleinen Trommelfells, der basalen Gehörknöchelchen und des einfachen und dünnen Kortex ein vielversprechendes Modell für das Gehör basaler Säugetiere darstellt.

Kooperation festigt die Kohäsion zwischen Tieren und ist damit ein prägender Faktor von Sozialsystemen. Das vorliegende Projekt zielt darauf ab, wie individualisierte Beziehungen, die sich in der Art und dem Umfang soziopositiver Interaktionen zwischen Dyaden widerspiegeln, das Sozialsystem bei einem Fledermausmodell, Carollia perspicillata, formen. Zunächst werden die Effekte der interagierenden Individuen, sowie des Geschlechts, auf das kooperative Verhalten von Fledermäusen innerhalb ihrer stabilen sozialen Gruppe untersucht. Der Effekt von Vertrautheit auf die Kooperation steht im Zentrum des zweiten Projektteils. Dazu werden Verhaltensexperimente zur sozialen Körperpflege, sowie zum Futterbetteln und -teilen durchgeführt. Es werden Unterschiede in der Art, Häufigkeit und Dauer von Interaktionen und der begleitenden vokalen Kommunikation zwischen miteinander vertrauten Individuen der gleichen sozialen Gruppe und einander unbekannten Individuen erwartet. Die Ergebnisse werden dazu beitragen, unser Verständnis für die Rolle der Kooperation zwischen Individuen für die Evolution von Sozialsystemen bei Fledermäusen zu vertiefen.

Neurons in the dorsal nucleus of the lateral lemniscus (DNLL) are relevant for binaural processing especially during reverberations. Their basic biophysical and synaptic properties have been documented, but are not understood on the cellular and molecular level. It remains so far unclear what voltage gated ion channels (VGIC) underly their functional specifications and how synaptic inputs are modulated. One specific question that will be answered his how different VGIC (potassium and sodium channels) interact to allow these neurons to generate high firing frequencies at high temporal precision. This biophysical phenotype indicates that these features are differently solved compared to other auditory brainstem neurons with similar tasks. Thus, this project aims to understand the moleculare organisaiton of the postsynaptic integration and spike generation mechanism. Since, the GABAergic DNLL neurons are reziprocally connected substantial GABA is liberated onto each neuron. GABA not only leads to fast inhibitory singalling but also is well-documented to generally be an important neuromodulator in the auditory brainstem. Therefore, the action of GABA mediated neuromodulation will be investigated by characterizing its effect on synaptic inputs and VGIC.

Bei Säugern nimmt mit der Kopfgröße auch die Größe des Gehirns und der Neurone zu. Eine Zunahme der Neuronengröße führt unweigerlich zu einer erhöhten Membrankapazität. Um die integrativen Leistungen der Neurone und die damit verbundene Funktion eines Nukleus invariant von der Hirngröße zu halten, müssen neuronale Eigenschaften wie der Eingangswiderstand, die dendritische Morphologie, die synaptische Leitfähigkeitsowie die Anzahl und Lage von Ionenkanälen und Synapsen proportional skalieren. Eine Grundvoraussetzung, um die zellulären Mechanismen einer solchen Skalierung zu untersuchen, ist eine evolutiv konservierte Neuronenpopulation die unabhängig von Kopf- und Gehirngröße die gleiche Funktion innerhalb eines Schaltkreises erfüllt. Mit den Neuronen des medialen Nukleus des Trapezkörpers (MNTB), welche an der binauralen Verarbeitung und der spektro-temporalen Integration in der aufsteigenden Hörbahn beteiligt sind, steht ein solcher Schaltkreis zur Verfügung. Die Quantifizierung der biophysikalischen und morphologischen zellulären Parameter von MNTB-Neuronen, ihrer synaptischen Eingangsgrößen sowie der Position von Ionenkanälen und Synapsen bei unterschiedlich großen Säugetierarten, wie zum Beispiel Etruskerspitzmäuse, Wüstenrennmäuse und Ratten, ermöglicht es uns, die Konsequenzen der hirngrößenabhängigen Neuronen-Skalierung zu verstehen. Die Ergebnisse dieser Untersuchungen werden in einem biophysikalischen Zellmodell münden, um ihre jeweilige funktionale Bedeutung zu verstehen. Darüber hinaus können wir diesen komparativen Ansatz nutzen, um die funktionelle Rolle der Dendriten von MNTB Neuronen, welche bisher weitgehend unbekannt ist, zu untersuchen. Insbesondere durch die quantitative Bestimmung des Einflusses von dendritischen synaptischen Eingängen auf die synaptische Latenz und den Erfolg der Aktionspotentialgenerierung, können wir ihre potenzielle Rolle bei der hochfrequenten Signalweiterleitung erfassen.

Etruscan shrews need to constantly hunt to meet their energy requirements; because of their small body size, their relative energy consumption is very large. To efficiently hunt in close range, these animals rely especially on tactile sensory cues. However, for long-range detection of prey these animals likely use their well-developed hearing. Due to their small body and head size, these animals are expected to hear in frqeuncy range above 5 kHz and therefore are an ideal model system to investigate high frequency hearing and adaptations to miniaturization of mammalian neuronal circuits. We attempt to describe their auditory brainstem strucutes by applying structural and functional markers. After the initial anatomical characterization we will use electrophysiological techniques to comparatively study the properties of adutiory brainstem neurons. Thereby we will focus on biophysical and synaptic size adaptations within conserved neuronal circuits.
