Ulcerative colitis (UC), characterized by the chronic inflammation of the colon, can extend to the terminal ileum through the influx of colonic content via an inflamed ileoceacal valve (1). Ample evidence from in vivo and in vitro studies confirm that under these circumstances the gut mucosa integrity and functions, carried by intestinal brush border membrane (BBM) enzymes, are altered and might contribute to the pathogenesis of UC (2). Although both the activities and expressions of BBM are decreased in UC, the mechanism(s) by which this occurs is not yet elucidated. The endoplasmic reticulum (ER) plays a crucial role in the synthesis, correct folding and sorting of proteins, and is the main calcium (Ca2+) reservoir in the cell (3, 4). The link between ER proper function and ER Ca2+ levels is tight. This stems from the fact that the activity of most ER chaperones, ensuring the proper folding of proteins transiting the ER, requires high Ca2+ levels (5). A chronic decrease in ER Ca2+ levels can, therefore, lead to misfolded proteins accumulation and eventually to ER stress. Unresolved ER stress leads to intestinal inflammation and is a general feature of UC epithelium (6). Despite the available data on the dysfunction of BBM in UC, until now there are no current attempts to address ER stress and Ca2+ in the observed dysfunction.

The objective of the proposed research is to study using in vitro and in vivo models, the role of Ca2+ signaling in ER stress, hallmark of UC, and how this is linked to the dysfunction of the intestinal epithelial cells.

Die Aufgabenstellung und Zielsetzung dieses Projektes ist, den Einfluss des Sertoli Zell (SC)-spezifischen Knockouts des Gap Junction-Proteins Connexin 43 auf die räumliche und zeitliche Expression verschiedener Tight Junction-Proteine (Claudin-3,-5 und-11 sowie Occludin) der von SC gebildeten Blut-Hoden-Schranke nachzuweisen. Hierbei werden Knockout-Tiere mit Wildtyp-Tieren vergleichen.

Die somatotrope Achse hat eine zentrale Bedeutung bei der Stoffwechseladaptation während der Trächtigkeit. Hierbei kommt es zur Entkopplung dieses endokrinen Systems welche mit einer spezifischen Reduktion des hepatischen Wachstumshormonrezeptors (GHR) einhergeht. Um detailliertere Erkentnisse über diesen physiologischen Anpassungsschritt zu gewinnen, ist Ziel des Projektes eine Sandwich-Kultur boviner Hepatozyten zu etablieren, die den GHR funktionell exprimieren.

Das Monitoring und die Zustandsbewertungen mariner Wirbeltiere dient als Grundlage für die Erfüllung der Natura 2000-Berichtspflichten und den regionalen Meeresübereinkommen OSPAR, der Helsinki-Konvention sowie der Meeresstrategie-Rahmenrichtlinie (MSRL). Die Berichtsanforderungen für die deutsche Nord- und Ostsee betreffen insbesondere Seevögel, marine Säugetiere und Fische. Als Voraussetzung für die Bewertungen sollen Datenerhebungen im langfristig angelegten Biodiversitätsmonitoring mit Schwerpunkt in der "Ausschließlichen Wirtschaftszone" (AWZ) erfolgen.
Das Aufgabenspektrum wird in fachspezifische Teilaufgaben gegliedert und von Nachunternehmen mit den jeweils erforderlichen Kompetenzen wissenschaftlich umgesetzt.
Das ITAW übernimmt das Monitoring und die Bewertung von Meeressäugetieren. Regelmäßig werden mittels standardisierter, fluggestützer Erfassungen Daten zur Verteilung und Dichte von marinen Säugetieren erhoben. Das Vorhaben greift auf in den zurückliegenden Jahren erstellten Untersuchungsergebnisse zurück und führt die Aufgaben mit erweiterten Parametern fort.
Die Ergebnisse des Vorhabens sollen dem Bundesamt für Naturschutz (BfN) als wissenschaftliche Basis für die Erfüllung der Berichtspflichten sowie weiterer Vollzugsaufgaben (z.B. Bewertung von anthropogenen Eingriffen) in der AWZ dienen.

Resultate:

Im Rahmen des Projekts TopSpace wurden von 2014 bis 2017 Schweinswalerfassungen der deutschen Nord und Ostsee durchgeführt. Dabei konnte die in den Vorjahren beobachteten Verteilungsmuster bestätigt werden, die Zahl an Schweinswalen wurde in den Sommermonaten auf durchschnittlich 35.000 Tieren geschätzt. Die FFH Gebiete Sylter Aussenriff und Borkum Riffgrund erwiesen sich auch während dieses Projekts als Kerngebiete der Schweinswale und bleiben die einzigen Gebiete in der deutschen Nordsee, in denen wir seit Jahren regelmößig eine überdurchschnittlich hohe Zahl von Mutter Kalb Paaren beobachten können. In der Ostsee rechnen wir im Sommer mit etwa 3.000 Tieren, vor allem das Gebiet um Fehmarn zeichnet sich durch Mutter Kalb Sichtungen und überdurchschnittlich hohe Dichten aus. The study wants to gain insights into the basic ecology (including phylogeography) of placozoans and better understand the factors that have fueled niche separation. We collect placozoans from different habitats in the field along with monitoring ecological parameters and analysing the genetic composition of field samples. With this information at hand we can address for the first time such fundamental issues as niche separation and habitat preferences, dispersal, and environmental adaptation in placozoans.

Als Alternative zur betäubungslosen Saugferkelkastration soll die Kastration unter Isoflurannarkose - kombiniert mit einer Metacambehandlung - in großen Ferkelerzeugerbetrieben (über 200 Sauen) auf ihre Praktikabilität, gesundheitliche Unbedenklichkeit für die Ferkel und das durchführende Personal und die Wirtschaftlichkeit geprüft werden.

Development of an improved vaccine for progressive control of Contagious Bovine Pleuropneumonia"

This proposal is part of an ambitious multi-stage process. The ultimate goal is to develop and make available better vaccine for contagious bovine pleuro pneumonia (CBPP), which are significantly better than those currently available. The data obtained as part of the validation of the improved tests will also enable existing epidemiological models of CBPP transmission to be enhanced. The improved epidemiological models will enable the formulation of new, more appropriate and evidence-based control policies. The main output will be the identification of pathogen molecules that could be used for designing more effective vaccines. That will be achieved by comparing the in vitro and in vivo proteome of MmmSC (2 dimensional electrophoresis and mass spectrometry) and by functional inhibition studies using a panel of monoclonal antibodies (MAbs).The project is carried out in Kenya, Germany and Namibia.

Resultate:

Overall this project was very successful: Within the project, we have generated knowledge, technologies and products, which will considerably contribute to the progressive control of CBPP.

Knowledge

Until now, very few virulence factors have been described for Mycoplasma mycoides, which lack the typical toxins and secretion systems of many other bacteria. Here, we provide evidence that the galactofuranose containing galactan, the polysaccharide coating of Mycoplasma mycoides subsp. capri and mycoides, indeed is a virulence factor. This information supports the further development of a polysaccharide conjugated vaccine.

Data generated within this project also supports the so far anecdotal evidence that CBPP infections indeed lead to lifelong immunity, at least for up to a year as we tested here. This shows that it will be possible to generate longer lasting immunity through improved future vaccines than the short lived immunity generated by the currently available vaccines.

We also show, through three different approaches (Precision Cut Lung Slice (PCLS) model system, in vivo immunosuppression trial as well as the in vitro cell assays, activities 1.2, 5.2 to 5.5, resp.) that the host may react to the pathogen in a counterproductive way, which leads to the hypothesis that severe and sometimes fatal outcome of CBPP is partly due to a misdirected host immune response.

Technologies

During the project, in vitro assays allowing the study of the interaction of mycoplasma with lung epithelial cells, macrophages and dendritic cells have been established.

A multiplex qPCR for the simultaneous detection of bovine cells and mycoplasmas of the Mycoplasma mycoides cluster was developed, allowing the quantification of both host and pathogen within one system.

An in vitro infection model using Precision cut lung slices (PCLS) was established to study host-pathogen interactions, bacterial adhesion, cell tropism, host specificity and cytotoxicity for Mmm and Mmc.

First steps to improve the CBPP challenge model have been initiated within this project, where repeated intranasal spraying of the infectious material replaced endotracheal intubation. This model needs further improvement, and the need for a robust challenge model has now been taken to a new level, with discussions including major international CBPP researchers.

Products

Last but not least, two products came out as a result of this project.

Through a consortium led by VIDO-InterVac and KALRO, a prototype subunit vaccine including proteins identified by ILRI resulted in filing a patent in North America for a CBPP vaccine.

A point-of-care lateral flow assay diagnostic tool has been developed, and a collaboration with a private sector company has been established to produce our previously developed cocktail ELISA.

-Der Einfluss des Tourismus auf die Habitatnutzung des Birkwildes soll anhand telemetrierter Birkhühner und der touristischen Aktivitäten im NSG Lüneburger Heide untersucht werden.
-Wie arrangieren sich saisonal und tageszeitlich die Birkhühner in den verschiedenen Lebensphasen (Balz, Brut, Kükenaufzucht, Winter) mit dem Touristen in dem gemeinsamen Lebensraum, welche negativen Beeinflussungen existieren und muss gegebenenfalls die Besucherlenkung optimiert werden?
-Mit Hilfe der Telemetrie wird die Habitatnutzung von Birkhühnern im NSG Lüneburger Heide im Jahresverlauf ermittelt. Insbesondere sollen die Balz-, Brut- und Aufzuchthabitate sowie die Winterlebensräume bestimmt werden. Darüber hinaus sind exakte Entfernungen der Aufenthaltsorte der Birkhühner zu den Wanderwegen und Waldrändern zu erfassen.
-Die touristische genutzten Wege und Plätze werden entsprechende ihrer saisonalen und tageszeitlichen Nutzung kartiert.
-Die Habitatnutzungsanalysen aus den Jahren 2011 - 2012 werden um weitere Daten ergänzt, sodass aussagekräftige Ergebnisse erzielt werden können.
-Die Bestimmung des Nahrungsspektrums adulter Birkhühner mittels Kotanalysen unterstützt die Bewertung der bevorzugten Birkhuhnhabitate.
-Dokumentation der Zwischenergebnisse zum 31.12. des Jahres und Publikation der Endergebnisse zum 31.03.2017

Beschreibung: Tiere unterschiedlicher Genetik (ROSS 308, Lohmann Dual, Hubbard, Lohmann Brown in zwei Altersstufen) werden identisch gefüttert und gehalten, wobei ein reduzierender Einfluss eines mit Laurinsäure versetzen Futters auf den Infektionsstatus der zuvor künstlich mit Campylobacter jejuni infizierten Tiere geprüft werden soll. Im Rahmen der Untersuchungen werden Leistungsparameter wie die tägliche Futteraufnahme, die täglichen Zunahmen und die Futterverwertung erfasst. Neben der täglichen Kontrolle und Dokumentation des klinischen Allgemeinzustandes wird die Fußballengesundheit beurteilt. Infektionsversuche mit Campylobacter jejuni sollen klären, ob unterschiedliche Fütterungsvarianten einen Einfluss auf die Ausbreitung einer experimentell herbeigeführten Infektion haben.
