Chronische Darmerkrankungen des Hundes sind häufig1 und auf Grund der multifaktoriellen Auslöser schwer zu behandeln. Die natürliche Darmflora, das intestinale Mikrobiom, trainiert das Immunsystem und scheint eine große Bedeutung in der Gesundheit des Darms sowie der Toleranz des Immunsystems zu haben. Bei chronischen Darmerkrankungen kommt es häufig zu einer Dysbiose, ein Ungleichgewicht der Darmflora.
Die Immunabwehr von Säugetieren ist ein komplexes System. Ein neu entdeckter Abwehrmechanismus der angeborenen Immunantwort sind extrazelluläre DNA-Netze. Sie werden vor allem von neutrophilen Granulozyten nach einer Stimulierung durch Botenstoffe des Körpers oder durch Krankheitserreger gebildet. Nach ihrer Aktivierung stoßen Neutrophile ihre DNA nach außen. Diese ist kombiniert mit antimikrobiellen Substanzen in der Lage Erreger zu binden, zu entschärfen und zu töten. Somit helfen diese DNA-Netze Infektionen im Körper einzugrenzen. Es wurden jedoch auch schädliche Wirkungen von DNA-Netzen bei einer unzureichenden Regulierung gezeigt. Bei ausgewählten Autoimmunerkrankungen (immunvermittelte, hämolytische Anämie), systemischen und lokalen bakteriellen Infektionen konnte beim Hund ein erhöhter Gehalt an DNA-Netzen nachgewiesen werden. Im Zusammenhang mit chronischen Darmerkrankungen ist es von großem Interesse, dass bei Mäusen und Menschen eine vermehrte Bildung von DNA-Netzen bei Kolitis nachgewiesen werden konnte.
Weiterhin kann man bei chronischen Darmerkrankungen einen Zusammenhang mit dem enteralen Mikrobiom ziehen. Inwieweit dieses von DNA-Netzen beeinflusst wird, ist derzeit ungeklärt.
Die Grundidee dieses Forschungsvorhabens ist aufzuklären, welche Rolle DNA-Netze bei chronisch-entzündlichen Darmerkrankungen und in der Immunabwehr gegen bakterielle Infektionserreger des Hundes spielen oder wie sie das enterale Mikrobiom beeinflussen. Das wiederum ermöglicht Rückschlüsse auf die Entstehung und die Entwicklung neuer Ansätze für gezielte Therapien der chronischen Darmerkrankung.

Project Details:
The equine recurrent uveitis (ERU) is the most important eye disease in horses, but the exact pathogenesis is still not fully understood. Furthermore, the recurrent uveitis is also found in human as well as almost all domesticated animals. Since autoreactive antibodies are detectedable in diseased horses, the classical ERU is considered as an autoimmune disease. The classical form of ERU is characterized by an acute serofibrinous and plasmacellular inflammation of the ciliary body, iris, choroidea and other eye structures. The disease often results into a chronic form by recurring intervallic relapses. The impulses lead to a progressive destruction of intraocular structures and finally lead to blindness.
Besides autoimmune processes, Leptospira infections are discussed to contribute to ERU in humans, horses, dogs and cattle. In over 60% of ERU-diseased horses Leptospira interrogans has been detected in the VBF using PCR or microscopic agglutination test (MAT). Additionally, the microbiological culture of Leptospira interrogans from VBF has been proven in further studies. Nevertheless, the role of the pathogen in the pathogenesis of ERU is still controversially discussed. Since the eye is protected via special barriers as one of the immune-privileged compartments in the body, the immigration of protective immune cells into the eye is highly regulated. A damage of the blood retina barrier is a typical pathological feature of ERU and often leads to a large number of migrating immune cells into the eye. The question arises whether the barrier is disrupted by infectious agents such as Leptospira, or whether the infection is a consequence of the immune-mediated pathogenesis of the ERU. These questions have not been fully clarified yet. However, irrespective of the Leptospira detection in ERU-diseased horses, the pathogenesis of ERU seems to be a highly complex altered immune reaction of the eye. Interestingly, in some populations an accumulation of different leukocyte populations as granulocytes can also be found in ERU patients, which have been histologically detected in the retina. However, especially the role of neutrophil granulocytes in ERU is still unclear and the focus of this project.

The aim was to create a course on the topic of "Basic training in the use of fluorescence microscopy" for students of biology and veterinary medicine via the documentation system in the TiHo Moodle platform. This module can be completed by the students in self-study. It is underpinned by texts, videos and subject-specific questions that must be answered after processing and viewing the videos. Essential parts of the content are also made available online as summarizing units as OER in the Twillo portal and are particularly suitable for students from natural science courses, pupils and trainees in (veterinary) medical professions.

Auf das Coronavirus trainierte Spürhunde werden in diesem Herbst bei mehreren Musikveranstaltungen eingesetzt und mit Antigen- und rtRT-PCR-Tests verglichen. Dies wird weitere Erkenntnisse darüber liefern, wie Corona-Spürhunde am besten bei der Detektion infizierten Menschen eingesetzt werden können.

Project Details:
Das im letzten Jahr neu entdeckte Coronavirus SARS-CoV-2 (Betacoronavirus, Coronavirdae) verursacht
aktuell eine globale Pandemie. Die Todesrate wird aufgrund aktueller Berechnungen zwischen 0,25 %
und 12% geschätzt (Wilson et al. 2020, Mizumoto et al. 2020). Bisher gibt es keine Prophylaxe oder
Therapie gegen die durch SARS-CoV-2 verursachte Erkrankung "COVID-19". Die Herkunft von SARS-CoV2 ist bisher unbekannt, allerdings werden Fledermäuse und Schuppentiere aktuell als Überträger
diskutiert (Tsan-Yuk Lam et al. 2020). Daher sind experimentelle Tiermodelle eine effiziente, sichere,
schnelle und somit unabdingbare Basis für die Entwicklung und Testung von präventiven und
therapeutischen Maßnahmen gegen SARS-CoV-2-Infektionen beim Menschen.
Frettchen gelten als geeignete Testmodelle für die Erforschung der Pathogenese und Entwicklung von
Impfstoffen gegen das 2003 entdeckte SARS-CoV-1. SARS-CoV-1 und -2 nutzen über das virale spike (S)
Protein den Wirtszellrezeptor ACE2 (Hoffmann et al. 2020), um Wirtszellen zu infizieren. Daher werden
Frettchen bereits für die Erforschung von SARS-CoV-2 eingesetzt (Callaway 2020). Phylogenetische
Analysen des ACE2 Rezeptors (Abb. 1, Anhang 1) zeigen, dass eine enge genetische Verwandtschaft
zwischen dem ACE2 Rezeptor von Menschen und Kaninchen vorliegt, und sich somit Frettchen und
insbesondere auch Kaninchen deutlich besser für die Forschung zum SARS-CoV-2 eignen als z.B. das
häufig eingesetzte Mausmodell. Zusätzlich werden Hamster als geeignetes Modell angesehen (Cohen,
2020). Darüber hinaus sind nicht-menschliche Primaten (NHP) für die Testung von Impfstoffen von
zentraler Bedeutung, da sie die Immunantwort des Menschen am besten widerspiegeln. Ausgewählte
Wirkstoffe und Impfstoffe, die sich als geeignet und sicher in den oben genannten Tiermodellen
erwiesen haben, sollen zur abschließenden Bewertung in NHP-Modellen getestet werden.
Beim Testen von Impfstoffkandidaten und Antikörperpräparaten in Humanstudien besteht wie bei
anderen Coronaviren das Risiko einer erhöhten Anfälligkeit für SARS-CoV-2-Infektionen, was bei einer
natürlichen Infektion zu einer schwerwiegenderen Erkrankung führt. Dies kann durch eine
antikörperabhängige Verstärkung, durch verzerrte Immunantworten (z.B. durch T-Zellen) oder durch
eine Kombination solcher Mechanismen verursacht werden. Hinweise für eine solche Verstärkung
wurden bereits für Impfstoffe gegen SARS und MERS erhalten und es muss sichergestellt werden, dass
eine vergleichbare Problematik nicht bei einem Impfstoff auftritt, der zur Bekämpfung der COVID-19-
Pandemie eingesetzt wird. Daher ist es von größter Bedeutung, Tiermodelle zu etablieren, die den normale und den therapeutisch
verbesserten Krankheitsverlauf von COVID-19 für eine optimale Risikominimierung nachbilden. Daher
sollen hier schnellstmöglich ab sofort experimentelle Testmodelle im Frettchen, Kaninchen und Hamster
in den BSL-3-Laboren und auch Tierstallungen am Research Center for Emerging Infections and Zoonoses
(RIZ), sowie mit nicht-menschliche Primaten (NHP) am Primatenzentrum in Göttingen etabliert werden,
um diese Modelle für die unmittelbare Testung von neuen antiviralen Strategien und
Impfstoffentwicklungen einzusetzen. Für die Durchführung dieser Tierversuche ist ein höchstmöglicher
Standard im Sinne des Tierschutzes und der Biosicherheit erforderlich. Die beteiligten Einrichtungen
verfügen über die fachliche Expertise und auch institutionelle Voraussetzung mit modernster
Gebäudetechnologien, um diesen Standards gerecht zu werden. Damit können diese Einrichtungen mit
der Etablierung dieser Tiermodelle eine Plattform für die regionale, aber auch nationale und
internationale Forschung liefern, um schnellstmöglich die Testung von Wirkstoffen und Impfstoffen
umzusetzen.

Project Details:
Trotz der intensiven Bemühungen die Tiergesundheit zu verbessern, erkranken Schweine
immer noch häufig an Pneumonien. Diese führen zu großen wirtschaftlichen Verlusten.
Ursachen können sowohl Viren als auch Bakterien sein. In den meisten Fällen handelt es
sich jedoch um komplexe Faktorenkrankheiten, an denen mehr als ein Erreger sowie die
komplexe Stressreaktion des Wirtes bzw. die Wirtsimmunantwort beteiligt sind.
Das Ziel dieser Studie ist es zu erforschen, welche Wirkung bakterielle Co-Infektionen und
Faktoren des Wirtes auf den Verlauf einer Pneumonie beim Schwein haben. Zunächst soll
das bakterielle Wachstumsverhalten in Co-Kulturen von Actinobacillus pleuropneumoniae,
Streptococcus suis und Haemophilus parasuis analysiert werden. Darauf aufbauend soll
dann der Einfluss von Wirtsfaktoren auf die Vermehrung und Virulenz von den Bakterien
untersucht werden. Als Wirtsfaktoren sollen Komponenten des angeborenen Immunsystems
sowie Stresshormone berücksichtigt werden. Als erste Zelle bei Infektionen wandern
neutrophile Granulozyten ein. Dabei stellt die Ausbildung von extrazellulären DNA-Netzen
einen wichtigen Abwehrmechanismus dar. Diese können Pathogene binden und durch die
Einlagerung von Komponenten wie Histonen und antimikrobiellen Peptiden (AMP) abtöten.
AMP spielen sowohl in DNA-Netzen eingelagert als auch durch direkte antimikrobielle
Wirkung eine Rolle. Zu den AMP gehören in der Familie der porcinen Cathelicidine z.B. PR-
39 und PMAP-37. Der Einfluss von AMP und DNA-Netzen auf die drei Pathogene soll in
Einzelkulturen vergleichend zu Ko-Kulturen untersucht werden. Zum anderen soll der
Einfluss von Kurzzeit- und Langzeitstress auf Bakterien, AMP Wirkung und DNA-Netze
durch die Inkubation mit Stresshormonen analysiert werden. Als Kurzzeitstresshormone
sollen Adrenalin und Noradrenalin und als Langzeitstresshormon Kortisol verwendet werden.
Alle Hormone gehören zu den Katecholaminen. Die erhobenen Daten sollen im Anschluss
an das beantragte Projekt für die Testung in einem Zellkultursystem der Lunge verwendet
werden. Dort soll das Zusammenspiel aller Komponenten für umfassende Rückschlüsse auf,
die in vivo Situation erforscht werden.

The worldwide occurrence of resistant bacteria limits the efficiency of antibiotic-based treatment concepts. Therefore, new promising therapeutic approaches are needed, such as the strengthening of the host's defense by stimulating the immune system. Since the complex host-pathogen interactions are still poorly understood, detailed knowledge is required to apply therapeutic strategies based on the innate immune system. However, animal-free in vitro model systems for infection and interaction studies as well as for drug screenings are only a real alternative if the results obtained can be reliably transferred to the in vivo situation. However, due to lack of complexity, incorrect cell differentiation status, and lack of physiological conditions, in vitro systems do not sufficiently accurately simulate the in vivo situation during infection or inflammatory response.
The project aims to characterize the innate immune response with focus on lung epithelial cells against respiratory bacteria such as the human pathogen Staphylococcus aureus or the zoonotic pathogen Streptococcus suis in vitro and ex vivo. The complexity of the infection model is significantly increased by physiologically relevant oxygen conditions (defined hypoxic conditions, <10% O2 by means of a hypoxia glove box) and by 3D co-cultivation of human and porcine pulmonary epithelial cells and neutrophils.
This study will help to minimize false negative results in screening potential protective immunomodulators or antibiotics that are found to be effective in vitro but ineffective in vivo. Increased complexity of the model system also allows the in vitro system to approach the in vivo situation and will help to reduce the number of animals.

S. aureus is a multifaceted commensal organism and a potentially harmful human pathogen. It can cause a broad spectrum of infections, which can involve any organ. Beside toxin mediated diseases, it can also play a role as an intracellular pathogen. In vivo studies concern the association of S. aureus and gastrointestinal disorders focussed on the colonization of the gastrointestinal tract by S. aureus, and on the influence of staphylococcal toxins. In this study the influence of S. aureus infections on the function of human intestinal cells were investigated in an in vitro colon carcinoma (Caco)-2 cell model. It could be shown that S. aureus Newman wild type (WT) and the mutant strain S. aureus Newman Äeap exhibiting a lack of the extracellular adherence protein (Eap), were able to invade day 7 post-confluent Caco-2 cells followed by intracellular survival, persistence and replication, even though substantial less S. aureus Newman Äeap bacteria were invasive. Staphylococcal infection did not induce any cytotoxic effect observed by a membrane integrity test showing lactate-dehydrogenase (LDH)-release of infected cells remained unchanged compared to uninfected cells. This was in addition visualized microscopically with LIVE/DEAD Viability/Cytotoxicity Kit for mammalian cells after infection with S. aureus Newman WT expressing the green fluorescence protein (GFP). However, the function of the infected cells was altered: a decrease in enriched specific activity of sucrose from the marker glycoprotein human sucrase-isomaltase (hSI) was observable in the apical membrane fraction 48h after infection. Specific SI-activity was increased in the basolateral membrane fraction as well as decreased in the apical membrane fraction with no alterations in the catalytic capacity. Results obtained from this study suggest that infections with S. aureus could influence the sorting of hSI, leading to intestinal disorders followed by symptoms like diarrhea. Further investigations based on these results will give new insights in S. aureus gut infections and the association with gastrointestinal diseases, which could possibly enable therapeutic steps because of shedding light on the causes of symptoms.

In Sudan, traditional medicinal plants continue to form the basis of rural medical care. This is due to the fact, that these medicines are easily available and cheap. In Sudanese folk medicine, some plants were claimed to have therapeutic values as for example Gum Arabic.
Gum Arabic is a dried exudate obtained from the branches and stems of Acacia senegal and closely related species. It is a complex polysaccharide of high molecular weight which contains neutral sugars as rhamnose, arabinose, and galactose; acids such as glucuronic acid; minerals such as calcium, magnesium, potassium, sodium, and phosphorous. In folk medicine, Gum Arabic has been reported to be used for the treatment of inflammation of the intestinal mucosa, and to cover inflamed surfaces.
A study conducted in Sudan has proven that the addition of Gum Arabic to rat?s diet has positive effect in lowering serum cholesterol and triacylglycerol (TAG) levels. Interestingly, drugs that are able to interfere with cholesterol synthesis as statins have been shown by us to induce antimicrobial activity of neutrophils against S. aureus, namely the formation of neutrophil extracellular traps (NETs). Thus, it may be hypothesized that medicinal plant extracts as Gum Arabic might have a protective effect on S. aureus infections by boosting antimicrobial neutrophil functions and thereby improve the outcome of an infection.
Since there is nothing known about the potential of Gum Arabic extracts to boost the host immune system, the goal of this study is to test Gum Arabic extracts for their ability to enhance the antimicrobial capabilities of phagocytes such as neutrophils as the first line of defense against invading pathogens.

Project Details:
An efficient, cost-effective test system will be established to test a large number of people as quickly as possible
(> 1,000/day). In three process steps i) Samples are taken in mobile test centers
(Throat swab according to the standard of the Robert Koch Institute), ii) processed (RNA isolation) and iii) evaluated by means of PCR analysis according to the Charité Berlin1 protocol. Positive results from the screening will be
diagnostically confirmed by certified laboratories and reported to the local health department. The mobile stations
are built on the basis of flexibly set up shipping containers and both for taking swabs
as well as the purification set up. In this way, a strict separation between the removal and
the processing of potentially infectious material in the mobile container and the subsequent analysis of the
non-infectious RNA samples secured in separate laboratories.
