Emerging infectious diseases keep arising and remain a major threat to human health. Most of the emerging infectious diseases are zoonotic and originate in wildlife animals. The increasing emergence of novel zoonoses is often attributed to land-use change and in particular to agricultural expansion. However, the interaction with the biotic and abiotic environment requires specific adaptations that have largely remained enigmatic. The Leibniz ResearchCampus EcoPaths will apply principles of evolutionary ecology to gain a causal understanding of the mechanisms that allow zoonotic pathogens to persist, multiply, and cycle in the environment outside of the human host. Through a highly integrative approach, ecological drivers of the evolution of Clostridium difficile, Enterococcus faecium / E. faecalis, as well as Porcine Coronaviruses will be studied. The three emerging groups of zoonotic pathogens feature different host ranges, genetic variability, and transmission rates, and hence are exemplary for different life strategies. The Leibniz ResearchCampus EcoPaths is the first consortium to merge the complementary expertises of the six institutions DSMZ, DPZ, TU Braunschweig, TiHo, Thünen Institute, and HZI in the fields of bacterial cultivation, comparative genomics, molecular evolution, prokaryotic biochemistry and regulation, molecular epidemiology, immunology and infection biology. This will enable the partners to closely collaborate in the elucidation of (1) the persistence and (2) cellular responses of pathogens in the environment, and (3) the role of the animal host as reservoir and source of environmental cycling, and to address particularly innovative aspects including the analysis of historic reservoirs based on ancient DNA, modelling the ecological niches of pathogens, determining in situ growth rates, and analysing biotic interactions of pathogens in microbiomes. As long-term objectives, EcoPaths strives to contribute towards the identification of the main pathogen reservoirs, to improve monitoring of pathogens and their transmission routes, and to aid in the development of mitigation strategies for zoonoses. EcoPaths will offer interdisciplinary training opportunities in a topical research field to its PhD students, and a dedicated, specific career development program for young postdoctoral researchers.

Therefore, we are particularly interested in this initial host-pathogen interaction that determines the outcome of the disease. In the present project, we will therefore investigate this important initial interaction of Mmm with the primary target cells of the host organism, both in vitro and in vivo, at the molecular level. In particular, we will elucidate the molecular basis of adhesion of the pathogen to the host cell. To this end, we will identify adhesins of Mmm and determine their cellular receptors. We will also study in detail the early response of host epithelial cells to contact with the pathogen. In addition, we will study the antibody response against molecules involved in the early phase of infection. The overall goal of this project is to better understand the molecular basis of the early interaction between pathogen and host. In doing so, we aim to identify target structures that may facilitate the development of new therapeutic tools.

Streptococcus suis is a frequent colonizer of the upper respiratory tract of pigs, but can also cause severe systemic diseases like meningitis and septicemia. However, pathogenesis of S. suis infection and the role of its virulence-associated factors, namely the pore-forming toxin suilysin (SLY) and the D-alanine-D-alanyl carrier ligase (DltA), is still not fully understood. So far, it is known that SLY damages different host cells by lytic pore formation and induces an inflammatory response leading to the release of cytokines, especially by monocytes/macrophages, and the recruitment of neutrophils. D-alanylation of lipoteichoic acids in the cell wall by DltA is known to increase resistance against antimicrobial peptides (AMPs) and phagocytosis of S. suis by neutrophils.
Our aim is to clarify the role of SLY and DltA in S. suis colonization of the porcine respiratory tract, systemic dissemination via the bloodstream, and invasion of the central nervous system. Thereby we will focus on their influence on innate immunity cells and the crosstalk between monocytes/macrophages and neutrophils. We hypothesize that the host compartments influence the expression of sly and dltA differentially and that vice versa SLY and DltA influence the host?s innate immune response by modulating the crosstalk between neutrophils and monocytes/macrophages contributing to the resistance of. S. suis towards immune defense mechanisms.
To investigate this, we will use complex in vitro cell culture systems that closely mimic the in vivo situation of the three main host compartments: a co-culture system of primary respiratory epithelial cells differentiated under air-liquid interface conditions and alveolar macrophages, porcine precision-cut lung slices, a reconstituted whole blood model, and the blood cerebrospinal fluid barrier model. Infection experiments will be performed with S. suis serotype 2 wild-type strain (wt), its isogenic mutants Δsly, ΔdltA, and ΔdltAΔsly as well as the respective complemented mutant strains.
We will start with the analysis of sly and dltA expression in the three compartments by quantitative real-time PCR and Western blotting, followed by the investigation of the neutrophil and the monocyte/macrophage response towards S. suis wt and sly- and dltA-deficient mutants. We will focus on the formation of neutrophil extracellular traps, reactive oxygen species production, phagocytic activity, and the release of certain cytokines and AMPs. Finally, we will investigate how secretions from monocytes/macrophages induced by S. suis infection influence neutrophils regarding their transmigration, inflammatory response, and phagocytic capacity, and vice versa.
The detailed investigation of the innate immune response towards S. suis infection in the three different compartments will help to better understand the switch of S. suis as a colonizer to an invasive pathogen, and how SLY and DltA are involved in this process.

Enteroinvasive bacterial pathogens are serious health threats for humans and animals as they can overcome the physical and innate immune barrier by the intestinal epithelium. There is evidence that such pathogens exploit host transcytotic pathways of different intestinal epithelial cell types that are otherwise used for antigen and nutrient uptake. Intestinal transcytosis and transcytosis pathways may considerably contribute to the disease outcome of infection. However, these processes are not fully understood. For example, it is not clear whether different pathogens share pathways for translocation. This project describes a research collaboration to clarify transcytosis of the enteroinvasive intracellular bacterial pathogens Salmonella enterica and Mycobacterium avium. Both Salmonella serovars and M. avium subspecies are known to use different intestinal cell types for entry into the host. However, translocation through the cells is largely unknown. The aim of this project is to define the mechanisms of translocation and the promoting factors by infecting intestinal epithelial organoids.

Bacterial diarrhoeal diseases in pigs are of great economic importance, affect animal welfare in all age groups and contribute significantly to the use of antibiotics in pig. Of the seven Brachyspira species found in pigs, most diarrhoeal diseases are triggered by the species Brachyspira hyodsenteriae (BHY) and Brachyspira pilosicoli (BPI). Treatment of diarrhoea caused by brachyspiral infections is very time-consuming and only a small number of microbial agents are approved for treatment. The significance of differences in the pathogenic potential of different bacterial isolates and the influence of the genetic background of the pig is still unknown. Therefore, a reassessment of the brachyspirial infection to improve the animal welfare and reduce the use of antibiotics is needed. Consequently, the aim of the present project is to understand the pathogenesis of Brachyspira using our established in vitro intestinal organoid model. In addition, the influence of different Brachyspira species and the influence of different genetic backgrounds of the pigs will be investigated.

Project Details:
As part of the SeroFaSt - Mhyo project, a serological on-farm rapid test for the detection of
Mycoplasma hyopneumoniae infections in pigs. This test is intended to
differentiate between vaccinated and infected animals directly on the farm,
without the need for time-consuming and cost-intensive laboratory tests. The rapid identification of
infected animals should enable immediate, targeted treatment and thus lead to greater animal welfare and
and better consumer protection.

Es wird allgemein angenommen, dass virale und bakterielle Co-Infektionen im Respirationstrakt prädisponierend für Infektionen durch Streptococcus suis sind. Wahrscheinlich beruhen die prädisponierenden Effekte auf einer Beeinträchtigung mukoziliärer Clearance- und Barrierefunktionen sowie auf Modulationen der Immunantwort, die Adhärenz und Invasion von S. suis fördern. Allerdings sind die Prozesse und Mechanismen der Interaktionen von S. suis mit co-infizierten respiratorischen Zellen bisher nur wenig bekannt. Unsere bisherigen Ergebnisse ergaben, dass porzine Influenzaviren (SIV) die Interaktionen von S. suis mit respiratorischen Zellen in einem zweistufigen Prozess beeinflussen. Initial wird die Adhärenz und Kolonisation der Streptokokken durch Bindung von Sialinsäureresten der Kapselpolysaccharide an Hämagglutinin an der Oberfläche SIV-infizierter Zellen vermittelt. In einem zweiten Schritt wird durch SIV-induzierte Ziliostase und Reduzierung der mukosalen Barriere die Invasion von S. suis in tiefere Zellschichten ermöglicht. Dieser Prozess ist sehr wichtig für die Pathogenität von S. suis, da er Voraussetzung für die weitere Invasion in den Blutkreislauf und die Ausbreitung des Erregers ist.
Im Folgeprojekt werden wir uns auf die Effekte mukosaler Schäden durch co-infizierende Pathogene auf Interaktionen von S. suis mit der respiratorischen Barriere konzentrieren. Wir gehen davon aus, dass das Ausmaß der mukosalen Schädigung einen entscheidenden Einfluss auf den Verlauf einer S. suis Infektion hat. Wir werden die gleichen primären Zellmodelle nutzen wie im vorherigen Projekt. Allerdings werden wir als co-infizierendes Pathogen statt SIV nun Bordetella bronchiseptica einsetzen. Von B. bronchiseptica ist bekannt, dass in Schweinen Co-Infektionen mit S. suis auftreten (beide sind am Porcine Respiratory Disease Complex beteiligt). Zudem haben experimentelle Infektionen gezeigt, dass B. bronchiseptica Schweine für intranasale Infektionen mit S. suis prädisponiert. Unsere bisherigen Untersuchungen haben außerdem ergeben, dass B. bronchiseptica mukosale Schäden verursacht, die von Ziliostase bis zur Ablösung Zilien-tragender Epithelzellen reichen, was zur Förderung der Adhärenz, Invasion und Zytotoxizität von S. suis führte. Daher sind unsere Hauptziele (i) die Charakterisierung von Effekten (milder und schwerer) mukosaler Schäden durch B. bronchiseptica auf die Adhärenz und Invasion von S. suis, (ii) die nähere Klärung der Rolle des S. suis Toxins Suilysin in diesen Interaktionen sowie (iii) die Analyse der Zellantwort von B. bronchiseptica-infizierten respiratorischen Zellen auf S. suis. Als Erkenntnisgewinn erwarten wir nähere Einblicke in die komplexen Pathogen-Wirt Interaktionen während der initialen Phase von S. suis Infektionen und, allgemein, während bakteriell-bakterieller Co-Infektionen im porzinen Respirationstrakt. Darüber hinaus können unsere Analysen der Wirtszellantworten dazu beitragen, Modulationen mukosaler Immunantworten durch Co-Infektionen zu identifizieren.

Mycobacterium (M.) avium ist neben M. bovis die wohl bedeutendste mykobakterielle Spezies mit tiermedizinischer Relevanz. Taxonomisch ist sie in vier Subspezies eingeteilt: M. avium subsp. avium (MAA), M. avium subsp. silvaticum (MAS), M. avium subsp. paratuberculosis (MAP) und M. avium subsp. hominissuis (MAH). MAA, MAS und MAP sind obligate Tierpathogene. MAA und MAS verursachen die Geflügeltuberkulose bzw. Geflügeltuberkulose-ähnliche Veränderungen in Wildtauben. MAP ist der Erreger der Paratuberkulose der Wiederkäuer. Dagegen ist MAH ein Umweltkeim und opportunistischer Erreger von Infektionen vornehmlich des Schweins und Menschen, der aber auch sporadisch bei anderen Tierspezies gefunden wird. Relativ wenig weiß man über die Pathogenität und Wirtspräferenz der einzelnen M. avium Subspezies. Trotz ihrer hohen genetischen Identität unterscheiden sie sich bezüglich ihrer Genomgröße und des Genomaufbaus. Es wird vermutet, dass die Subspezies-spezifischen Gene und Genomabschnitte für Eigenschaften codieren, die zu Unterschieden in der Pathogenität und Wirtsanpassung beitragen.
Seit langem ist bekannt, dass MAA Stämme nach Infektion der Maus virulenter sind als MAH oder MAP Stämme. Eigene Untersuchungen zeigen, dass zur gesteigerten Virulenz von MAA eine nur bei MAA infizierten Mäusen vorkommende Bildung von MAA beherbergenden, Stickstoffmonoxid produzierenden, monozytären, myeloiden Suppressorzellen (mMDSC) beiträgt. Diese Zellen beeinflussen die Immunantwort der Maus und tragen zur Verschlechterung des Infektionsverlaufs bei. Im beantragten Projekt soll in einem systembiologischen Ansatz ermittelt werden, welche Eigenschaften von MAA zur Bildung von mMDSC und zum Überleben in mMDSC beitragen. Dies soll über die Analyse des in vivo Transkriptoms und Proteoms in der Maus, kombiniert mit Daten von Mausinfektionen, mit einer Transposonbank von MAA durchgeführt werden. Wir erwarten uns Erkenntnisse zu den Grundlagen der M. avium Pathogenität und Virulenz, die zur Vorbeugung, Erkennung oder Behandlung von M. avium Infektionen und auch anderer mykobakterieller Infektionen bei Tier und Mensch nutzbar wären.
