Ziel des Projekts ist es, erstmals einen Beitrag zur Aufklärung der vokalen Flexibilität eines vokal aktiven und stammesgeschichtlich basalen Primaten während der Entwicklung zu leisten. Unsere bisherigen Forschungsergebnisse an der basalen Primatenart Mausmaki deuten auf eine unerwartet hohe vokale Flexibilität in der frühen Ontogenese, die in der weiteren Entwicklung abnimmt. Zu ihrer Untersuchung schlagen wir drei verschiedene, miteinander verzahnte Arbeitsprogramme vor. Die Präsenz und Rolle von Brabbeln (babbling) und vokalem Austausch (vocal-turn taking) während der Entwicklung sowie potentielle Determinanten, die zu entwicklungsbedingten Veränderungen in der vokalen Struktur führen, wollen wir durch standardisierte bioakustische und videografische verhaltensbiologische Untersuchungen an einem Datensatz von insgesamt 26 Kindern währen der Entwicklung und ihren 13 Müttern erfassen. Über Maschinenlernansätze, die auch selbst-organisierende neurale Netze einschließen, wollen wir die Dynamik der Veränderungen während der Entwicklung automatisiert quantifizieren und mögliche, zu Grunde liegende Regeln ableiten. Über Larnyxexperimente, verbunden mit histologischen und MikroCT-Untersuchungen des Vokaltrakts sowie über Hochgeschwindigkeitsvideoaufnahmen von Mund- und Zungenbewegungen während des Vokalisierens, wollen wir einen ersten Einblick in die Mechanik der Lautproduktion eines basalen Primaten erhalten und mögliche biomechanische Zwänge der vokalen Flexibilität erfassen. Die integrative Bearbeitung der verschiedenen Datensets wird eine Validierung der eingesetzten Methoden erlauben. Das Vorhaben stellt eine wichtige empirische Grundlage für die vergleichende akustische Kommunikationsforschung bei Säugetieren dar und trägt dazu bei, die stammesgeschichtlichen Wurzeln der vokalen Flexibilität in der Primatenkommunikation als wichtigen Baustein für die Evolution von Sprache zu verstehen.

The project aims to employ modern genetic tools to identify lemur populations of high conservation value to help prioritizing conservation efforts and to develop and implement conservation strategy for the most threatened and valuable lemur populations. The following points constitute the major project objectives: Objective 1: Identify and sample suitable populations of two lemur model species in the study region. Objective 2: Characterize the populations with regard to their genetic diversity, genetic uniqueness, and genetic connectivity to identify the site(s) with the highest conservation value and concern.

The global biodiversity crisis that is affecting ecosystems worldwide is a major subject of concern and is expected to worsen with ongoing global changes. Climate change together with other anthropogenic factors will lead to the displacement of many favorable environments in the next decades owing to fast altitudinal and latitudinal shifts, with greatest predicted impacts in biodiversity hotspots. These shifts will lead to an increase in Habitat Loss and Fragmentation (HL&F), the main threats to Biodiversity worldwide. There is therefore a need to understand the consequences of HL&F, and to identify the barriers to gene flow at various spatial and temporal scales. We study HL&F in the context of past and future environmental changes, across taxa and regions. We use Madagascar as a model region and use comparative genomic analyses to infer generalizable critical features of ecological networks across various taxonomic and spatial scales. In particular, we identify barriers to gene flow based on genomic datasets and study the effects of forest fragmentation on the vegetation, and the abundance and parasite load of two mouse lemur and two rodent species.

Marked climatic oscillations between glacial and interglacial periods had worldwide consequences for vegetation as well as animal population dynamics. The importance of these shallow-time (on geological and evolutionary timescales) geographic dynamics for shaping current biodiversity and biogeography patterns is increasingly stressed, although rarely analyzed in an innovative integrated manner. One of the necessary steps in order to understand the drivers of biodiversity is to synergize the efforts from various research fields by, for example, reconstructing the interplay between the degree and frequency of historic forest cover changes and demographic changes of forest-dependent organisms. This study aims to integrate validated records of vegetation and climate dynamics with inferred population dynamics to reconstruct the dynamics of forest landscapes and of populations of forest dwelling species over space and time in a primate model endemic to Madagascar. Madagascar developed a unique biodiversity during its long isolated history. Despite the long-lasting interest in the natural history of the island, much is still unknown about the biodiversity dynamics and long-term ecology of this continental island. This multidisciplinary project aims to integrate demographic inferences based on molecular datasets of mouse lemurs with validated high resolution vegetation dynamics based on paleoecological reconstructions obtained from the same study sites reaching back to the Last Glacial Maximum (LGM). To reach these goals, study sites in northwestern and northern Madagascar were visited for the joint collection of (paleo)ecological and population datasets and samples of mouse lemurs in direct vicinity to each other. For the paleoecological part sediment cores from lakes were drilled and complemented with samples of modern pollen rain and vegetation data. The sediment cores are subjected to temporal high-resolution pollen and charcoal analyses, radiocarbon dating and multivariate modelling of the vegetation and climate dynamics over time and space. The lemur samples are analyzed by applying RADSeq and NextSeq sequencing techniques on various subsets of samples. This study will contribute substantially to a deeper understanding of the evolutionary history and future prospects of lemur populations in view of ongoing habitat fragmentation and future climate change.

The development of effective conservation measures for endangered species requires a detailed knowledge on the remaining individual numbers, the distribution of species and the factors causing potential threats to their survival. Moreover, it is important to understand the degree of intra-specific differentiation (e.g. phenotypic, ecological, genetic) that can be the result of rather recent (anthropogenic) or ancient evolutionary processes (barriers, landscape). The aim of this project is to understand the population structure of different lemur species across their distribution in view of drastic anthropogenic disturbances. Ancient and recent processes of genetic differentiation shall be identified in order to develop effective conservation measures. With this analysis, we will gain important insights into the evolutionarily significant units as well as and the management units of these threatened primates that will be needed to formulate long-term management plans.

A central question in evolutionary and conservation biology is how and to what extent species adapt to habitat fluctuations in their natural environment and to what extent environmental flexibility is linked to distribution and speciation. We address these questions in a longterm research programme by a comparative and integrative approach to nocturnal lemurs, a unique model for basal primates. Lemurs are endemic to Madagascar. Current research revealed a much higher species diversity in nocturnal lemurs than previously assumed. Lemurs underwent a broad-scale adaptive radiation after their initial colonization of Madagascar about 60 Mya. Extant lemurs consist of more than 100 species which inhabit the various forest habitats of the Island. Many species, in particular nocturnal species, have only been described recently, partly by our group. The underlying colonisation history and the factors driving this diversification are so far only poorly known. Here, we aim to understand the factors that promoted this radiation and shaped distribution, adaptation and genetic structures of lemurs. We are also addressing the question of the basis of reproductive isolation of species. These analyses are focused on the genera Microcebus spp., Lepilemur spp. and Avahi ssp. Whereas some species within a taxon show quite limited, others show broad distributions. Nocturnal lemurs belong to the endangered primates due to anthopogenically caused habitat degradation and poaching. We perform field studies, supplemented by experimental studies, particulary in north-western, northern and eastern Madagascar, using comparable methods, to study the ecology, behaviour, communication, parasite infections and the genetics of populations of lemurs of the same taxon showing contrasting distribution and to assess sensory, vocal, ecological and social determinants of species flexibility and its link to genetics. Our current research focus is on mouse lemurs, sportive lemurs and woolly lemurs. Results will deliver important tools to monitor and manage the biodiversity in tropical forests. Furthermore they will be a first step to understand the evolution of the unique species diversity of lemurs in Madagascar and the origin and evolution of our own primate ancestry.

The Ankarafantsika National Park and the Mariarano forest contain different habitat types, including xerophytic forests, dry deciduous forests, semi-humid gallery forests along rivers or lake shores, coastal mangrove forests (Mariarano) and secondary vegetation formations that may develop after bushfires or selective wood extraction. This mosaic of habitat types offers many different ecological niches for lemurs and other forest dwelling organisms. Both forests are unique in northwestern Madagascar by harboring five endangered or critically endangered lemur species, one of which, the golden brown mouse lemur (Microcebus ravelobensis) was only recently described by our group in 1998. Moreover, the Ankarafantsika National Park comprises the largest remaining continuous patch of dry deciduous forest in northwestern Madagascar and is therefore of utmost importance for the longterm preservation of biodiversity in this biogeographic zone. Knowledge on the environmental flexibility of lemurs is still in its infancy, but urgently needed for conservation management. The aim of this longterm programme is to study the conservation biology of these lemur species, including their vulnerability towards diseases, at sites, for which we already have basic data. This knowledge will help to understand the environmental flexibility of species, how macrogeographic events such as climate change affect lemur?s life history and long-term survival and provide empirical data for long-term conservation management of lemurs in northwestern Madagascar.

Ein vokales Repertoire enthält eine artspezifische Liste von Lautäußerungen, die von den Tieren produziert werden. Diese Listen bilden die methodische und theoretische Grundlage für eine Reihe von Forschungsfragen verschiedener Forschungsdisziplinen. Dennoch gibt es keine Einigkeit darüber, wie ein vokales Repertoire zu erstellen ist oder wie Ruftypen zu definieren sind. In den meisten Studien wurden die Ruftypen durch visuelle Klassifizierung anhand von Spektrogrammen bestimmt, die durch statistische Analysen auf der Grundlage der gemessenen akustischen Merkmale einer Vokalisation bestätigt wurden. In neueren Veröffentlichungen wurden objektivere mathematische Ansätze wie "supervised"" oder ""unsupervised"" Clustering-Algorithmen entwickelt. Allerdings erfordern auch diese Algorithmen oft eine Vorklassifizierung des Beobachters, entweder, weil eine Anzahl von erwarteten Clustern bestimmt werden muss, oder, weil das Clustering-Ergebnis auf seine biologische Plausibilität hin überprüft werden muss. Beide Methoden können zu einer Überschätzung der Anzahl von Ruftypen führen, insbesondere bei Arten mit sehr variablen Vokalisationen oder bei Ruftypen mit graduellen Übergängen. Eine akustische Variation allein reicht also nicht aus, um eine Verhaltensrelevanz widerzuspiegeln. Dies erfordert auch eine Kontextspezifität der Rufe und die Fähigkeit des Gehirns, die akustische Variationen wahrzunehmen und verarbeiten zu können. In diesem Projekt erstellen wir vokal Repertoire, indem wir die mathematische Klassifizierung der Vokalisationen mit den Verhaltenskontexten, in denen die Rufe produziert wurden, sowie mit den physikalischen Hörfähigkeiten (z. B. Hörbereich, Zeit- und Frequenzauflösung) der jeweiligen Art kombinieren. Wir verwenden diesen Ansatz für zwei kleine Säugetierarten: die mongolische Wüstenrennmaus, ein wichtiges Tiermodell in der Hörforschung, und die Etruskerspitzmaus, die aufgrund ihres kleinen Trommelfells, der basalen Gehörknöchelchen und des einfachen und dünnen Kortex ein vielversprechendes Modell für das Gehör basaler Säugetiere darstellt."

Bei Säugern nimmt mit der Kopfgröße auch die Größe des Gehirns und der Neurone zu. Eine Zunahme der Neuronengröße führt unweigerlich zu einer erhöhten Membrankapazität. Um die integrativen Leistungen der Neurone und die damit verbundene Funktion eines Nukleus invariant von der Hirngröße zu halten, müssen neuronale Eigenschaften wie der Eingangswiderstand, die dendritische Morphologie, die synaptische Leitfähigkeitsowie die Anzahl und Lage von Ionenkanälen und Synapsen proportional skalieren. Eine Grundvoraussetzung, um die zellulären Mechanismen einer solchen Skalierung zu untersuchen, ist eine evolutiv konservierte Neuronenpopulation die unabhängig von Kopf- und Gehirngröße die gleiche Funktion innerhalb eines Schaltkreises erfüllt. Mit den Neuronen des medialen Nukleus des Trapezkörpers (MNTB), welche an der binauralen Verarbeitung und der spektro-temporalen Integration in der aufsteigenden Hörbahn beteiligt sind, steht ein solcher Schaltkreis zur Verfügung. Die Quantifizierung der biophysikalischen und morphologischen zellulären Parameter von MNTB-Neuronen, ihrer synaptischen Eingangsgrößen sowie der Position von Ionenkanälen und Synapsen bei unterschiedlich großen Säugetierarten, wie zum Beispiel Etruskerspitzmäuse, Wüstenrennmäuse und Ratten, ermöglicht es uns, die Konsequenzen der hirngrößenabhängigen Neuronen-Skalierung zu verstehen. Die Ergebnisse dieser Untersuchungen werden in einem biophysikalischen Zellmodell münden, um ihre jeweilige funktionale Bedeutung zu verstehen. Darüber hinaus können wir diesen komparativen Ansatz nutzen, um die funktionelle Rolle der Dendriten von MNTB Neuronen, welche bisher weitgehend unbekannt ist, zu untersuchen. Insbesondere durch die quantitative Bestimmung des Einflusses von dendritischen synaptischen Eingängen auf die synaptische Latenz und den Erfolg der Aktionspotentialgenerierung, können wir ihre potenzielle Rolle bei der hochfrequenten Signalweiterleitung erfassen.

Kooperation festigt die Kohäsion zwischen Tieren und ist damit ein prägender Faktor von Sozialsystemen. Das vorliegende Projekt zielt darauf ab, wie individualisierte Beziehungen, die sich in der Art und dem Umfang soziopositiver Interaktionen zwischen Dyaden widerspiegeln, das Sozialsystem bei einem Fledermausmodell, Carollia perspicillata, formen. Zunächst werden die Effekte der interagierenden Individuen, sowie des Geschlechts, auf das kooperative Verhalten von Fledermäusen innerhalb ihrer stabilen sozialen Gruppe untersucht. Der Effekt von Vertrautheit auf die Kooperation steht im Zentrum des zweiten Projektteils. Dazu werden Verhaltensexperimente zur sozialen Körperpflege, sowie zum Futterbetteln und -teilen durchgeführt. Es werden Unterschiede in der Art, Häufigkeit und Dauer von Interaktionen und der begleitenden vokalen Kommunikation zwischen miteinander vertrauten Individuen der gleichen sozialen Gruppe und einander unbekannten Individuen erwartet. Die Ergebnisse werden dazu beitragen, unser Verständnis für die Rolle der Kooperation zwischen Individuen für die Evolution von Sozialsystemen bei Fledermäusen zu vertiefen.
